Objective: To explore whether there exists significant difference in DNA methylation of glucose transporter 4(GLUT4) gene between subjects with and without ostructive sleep apnea syndrome(OSAS) and potential relationship of DNA methylation of GLUT4 and OSAS-related parameters.Methods: 14 Han Chinese hypertensive males, aged between 30-50 years, with severe OSAS confirmed by polysomnography and with normal hepatic and renal function were consecutively selected as the OSAS group and 16 age-, BMI-, mean systolic and diastolic blood pressure-matched Han Chinese hypertensive males without OSAS were selected as the control group from the hypertension department of Xinjiang Uygur Autonomous Region between Jan to March 2010. DNA methylation of GLUT4 gene was assessed using Mass ARRAY. After high methylation sites were found, they were analyzed in case group, comprised of 66 subjects with moderate/severe hypoxemia cases and control group comprised of 49 subjects.Results: 1.This experiment was tested in a total of 30 samples; 4 Cp G island was detected in each sample and 86 Cp G sites, T cut into 56 Cp G units(each unit of Cp G containing single or multiple Cp G sites), analysis rate was 78.6%. The highest methylation rate was 45%, the methylation rate less than or equal to 10% accounted for 88.9%, the methylation rates more than 10% accounted for 11.1%, and the overall methylation level was lower. DNA methylation level was higher in OSAS group than was the non-OSAS group, whereas it did not reach statistical significance. 2. DNA methylation level of GLUT4 gene in all Cp G showed no significant differences between OSAS and control group, whereas DNA methylation of the locus2.10 and locus2.14 were observed to be significantly higher in OSAS subjects than in the control group with normal FBG(loc2.10[0.5(0,3.0) vs 3.0(2.0,6.5),p=0.014] 、 loc2.14[1.5(0,4.0) vs 6.0(2.0,9.5),p=0.004], respectively). 3. Spearman correlation analysis showed that lowest saturation of oxygen was in a significant negative correlation with the DNA methylation level of the locus2.10 and 2.14(r=-0.46, p=0.027 and r=-0.51, p=0.004, respectively).DNA methylation of the locus2.14 was significantly positively correlated to the serum concentration of IL-1β(r=0.499,p=0.018). 4. in severe OSAS hypermethylation of sites loc2.10 [4.5(2.0,23.0) vs 9.0(4.5,55.0)], loc2.14[6.0 2.0,24.0 vs 13.5(6.0,40.3)] and further found that hypoxemia group high methylation sites. Loc2.3[9.0(6.0,11.0) vs 10.5(8.0,18.5)], loc2.6[5.0 4.0,7.0 vs 6.5(5.0,49.3)], loc2.13[17.0 14.0,28.0 vs 2.7(16.0,80.0)], loc2.17[6.0 1.0,14.0 vs 18.0(3.5,67.5)], P < 0.05, the difference has statistical significance.Conclusions: otherwise healthy OSAS males exhbited higher DNA methylation levels of some of the locus, particularly locus 2.10 and 2.14, of GLUT4 gene,and related With hypoxia and inflammatory biomarkers.
