OBJECTIVE: To build the alveolar ridge defect model of Beagles, compare this new biological bone materials and Bio-Oss bone repair bone defects When the discussion and analysis on the use of barrier films with and without a barrier membrane for guiding bone to distinguish the effect of providing some guidance for clinical use.METHODS: 6 local healthy adult beagles were selected, without limitation on the gender.. Before surgery, the third and fourth premolar teeth, and the first molar of mandible in both sides were removed. After 3 months of natural healing, 2 three-wall box-shaped defect models [size: 9mm(length) * 7mm(width) * 5mm(height)] were made in mandible agomphosis area of both sides, and the interval between 2 defects in the same side was 5mm. 4 defects were treated randomly, and divided into Bio-Gide membrane with Bio-Oss bone group, and Bio-Gide membrane with biological bone material group,and biological bone material group,and blank control group,The beagles in corresponding group were put to death after 1 week,4 weeks, 8 weeks, 12 weeks and 24 weeks of surgery respectively. The general observation, spiral CT check, and three-dimensional reconstruction observation through Simplant 16.0 were carried out. Besides, the statistical analysis was made on the achieved data.RESULTS:1. Spiral CT check and CT value measurement. At different time points, the CT value of the Bio-Gide membrane with Bio-Oss bone group was close to that of the Bio-Gide membrane with biological bone material group. The difference from their comparison did not have statistical significance(P>0.05). The CT value of the biological bone material group was lower than that of the above 2 groups. The difference from the pairwise comparison had statistical significance(p<0.05). CT value of the blank control group was the lowest. Compared with other groups, the obvious difference had statistical significance(p<0.05).2. Three-dimensional reconstruction observation, bone volume and bone absorption rate measuring.Three-dimensional reconstruction observation meets the features of the CT observation. After one week, the bone volume of the bone defect area in Bio-Gide membrane with Bio-Oss bone group, Bio-Gide membrane with biological bone material group and biological bone material group was similar to each other. The difference did not have statistical significance(P>0.05). The blank control group showed the lowest bone volume. Compared with other groups, the difference was significant(p<0.05). After 4 weeks, 8 weeks,12 weeks,24 weeks, the comparison between the Bio-Gide membrane with Bio-Oss bone group and the Bio-Gide membrane with biological bone material group did not have difference(P>0.05). The value of the biological material group was lower than the above two groups. After pairwise comparison, there was some difference(p<0.05). The blank control group showed the lowest value, and it had difference compared with other groups(p<0.05). Though the calculation of the bone absorption rate, it turns out that the difference from the comparison between the Bio-Gide membrane with Bio-Oss bone group and the Bio-Gide membrane with biological bone material group did not have statistical significance at the same time points(P>0.05). Through the pairwise comparison for other groups, the difference was significant(p<0.05). Moreover, after the statistical analysis was carried out at these 4 time points, it showed that there was great difference between 12-week and 24-week bone absorption rate(p<0.05).CONCLUSION: 1. This new biological bone material has function of certain bone guidance and capability of repairing the bone defect, it’s effects similar to Bio-Oss bone.2. This new biological bone material can achieve better osteogenesis effect when it is using together with the Bio-Gide membrane than using alone.
