Objective To observe the effect of different doses of Apostichopus japonicus glycosaminoglycan(HGAG) on the proliferation of peripheral blood mononuclear cells(PBMC) and the expression of CD80、CD86 of healthy people and Lung cancer patients in vitro and the clinical significance.Methods 40 cases diagnosed as lung cancer patients clinically in Affiliated Hospital of Qingdao University Respiratory(west coast of Qingdao City Hospital Respiratory) and40 cases of healthy people in health check centers were collected during October 2014 to May 2015, PBMC extracted from peripheral blood of healthy people and Lung cancer patients with ficoll-hypaque density gradient centrifugation were incubated with different doses of HGAG in vitro for 24 hours. The proliferation activity of PBMC was tested by CCK-8 assay, the expression of CD80 and CD86 molecular on lymphocyte was detected by flow cytometry.Results(1) HGAG can accelerate the proliferation of PBMC of healthy people obviously within the range of 1~50μg/ml compared with the control group(p<0.05),especially at10μg/ml doses group(p<0.01), while, HGAG can not improve the proliferation of PBMC of healthy people at 100μg/ml doses group(p>0.05);(2) HGAG can increase the proliferation of PBMC of Lung cancer patients obviously within the range of10~100μg/ml and with doses-dependent manner compared with the control group(p<0.01).(3)When the concentration of HGAG increased, the proliferation of PBMC of healthy people decreased; while, the proliferation of PBMC of Lung cancer patients increased within the range of 0.1 ~ 100μg/ml.(4)The expression of CD80 and CD86 on peripheral blood lymphocytes of lung cancer patients was significantly lower than healthy people, and the difference was statistically significant(P <0.01).(5)HGAG can significantly promote the expression of CD80 molecules on peripheral blood lymphocytes of healthy people within the range of 1 ~ 50μg / ml, as compared with the control group,the difference was statistically significant(P <0.05), and the effect of 50μg / ml group performance better than 1μg / ml group and 10μg / ml group(P <0.05), there is a certain concentration dependent; HGAG can significantly promote the expression of CD86 molecules on peripheral blood lymphocytes of healthy people in 10 μg/ml group and 50μg/ml, as compared with the control group, the difference was statistically significant(P<0.05), and the effect of 50μg/ml group and 10μg/ml group is better than 1μg/ml group(P<0.05).(6)HGAG can significantly promote the expression of CD80 and CD86 molecules on peripheral blood lymphocytes of lung cancer patients in 10 μg/ml group and 50 μg/ml,as compared with the control group, the difference was statistically significant(P <0.05),and the effect of 50μg/ml group is better than 1μg/ml group and 10μg/ml group(P<0.05),and the effect of 10μg/ml group is better than 1μg/ml group(P <0.05).Conclusion HGAG can accelerate the proliferation of PBMC of lung cancer patients and healthy people and promote the expression of CD80 and CD86 molecules on peripheral blood lymphocytes of lung cancer patients and healthy people within a certain range of concentrations, which can promote the activation of T lymphocyte and improve immune function and anti-tumor effect.
