The Expression Of Co-Stimulater CD80 And CD86 In Cervical Premalignant And Malignant Lesions

OBJECTIVETo observe the expression of CD80 and CD86 in cervical premalignant and malignant lesions,as to get some information about the function of costimulatory pathy in the tumor immunity of cervical cancer.METHODSControlled with normal tissues,immunohistochemistry method was used to assess the expression of CD80 and CD86 in cervical cancer.3-μm sections were cut from paraffin blocks and mounted on polylysine-coated slides.After deparaffinization in xylene and rehydration in graded ethanols,heat induced epitope retrieval(HIER) was performed using a microwaveable vessel where the slides were immersed in Trisodium citrate Retrieval Solution and boiled for 15 minutes at 98℃in microwave oven.5-μm sections were also cut from frozen tissue on the cryostat and placed on polylysine-coated slides.Sections were air-dried and then slides were fixed for 10 minutes in acetone at -20℃.After pretreatment above,all sections were stained with CD80 and CD86 using Histostain-Plus kit.The specificity of the reaction was confirmed by use of substitution control in which PBS was used instead of the primary antibody.Positive control slides were included in each run and examined for appropriate staining. RESULTS1.The expression of CD80 antigen in different groups of cervical tissue:CD80 expressed on all the CINⅠ,CINⅡ,GINⅢand cervical cancer tissue,accompany by lymphocytes infiltration.CD80 antigen exists on the cancer nests and surrounding of necrotic area in cervical cancer tissue.There is only scattered CD80 in peri-cancerous tissue and normal cervical tissue which is away from cancer tissue.CD80 expressed on cytoplasm and nucleolus,but little even none expressed on cell membrane in cancer tissue.The expression of CD80 in cervical cancer tissue is lower than peri-cancerous tissue and normal cervical tissue and decreased progressively with the grades of cervical lesion.There is no significant difference of the CD80 antigen expression between peri-cancerous tissue and normal cervical tissue.2.The expression of CD86 antigen in different groups of cervical tissue:CD86 antigen also has two expression mode of cytoplast and nucleolus in cervical cancer tissue,peri-cancerous tissue and normal cervical tissue.The location and distribution are similar with CD80.The expression of CD86 in cervical cancer tissue is lower than peri-cancerous tissue and normal cervical tissue and also decreased progressively with the grades of cervical lesion.CONCLUSIONS1.The expression of CD80 and CD86 in cervical cancer tissue is lower than normal cervical tissue and decreased progressively with the grades of cervical lesion. The defected expression of CD80 and CD86 would lead to the tumor cell escaping the attack of immune system.2.CD80 and CD86 expressed on cytoplasm and nucleolus,but little even none expressed on cell membrane in cancer tissue,which indicated that tumor cell may have function failure of transfering CD80 and CD86 to cell membrane.This may correlated with the tumorigenisis and development of cervial cancer.
