The Study On The Efficacy Of Entiana Based On Di- Fferent Herb Pairs Treating Wind Musculosketal Rat Of RA As Well As Characteristic Spectrum

Objective: To study the compatibility of different drugs on gentian wind musculoskeletal RA rat efficacy and characteristic spectrum relationship, through wind musculoskeletal inducing rats,we try to establish rat model of RA and observe single herb gentian,gentian- clematis, gentian-mistletoe and gentian-tetrandra,which had an impact on routine blood indexes and ESR in RA rat blood,at the same time,the changes of contents of index components including gentiopicroside and swertiapatens in herb pairs’ decoction based on gentian were studied by high performance liquid chromatography in order to observe that herb pairs had an effect on characteristic spectrum about gentian.Method: Using type Ⅱcollagen and additional environmental factors called wind cold dampness,we tried to build a kind of rat model of rheumatiod arthritis in line with traditional Chinese medicine “arthralgia” feature called wind musculoskeletal disease and syndrome.A general status,weight changes,arthritis, routine blood and erythrocyte sedimentation rate about model rats were tested in this research.At the same time, the peak area of indicative chemical composition called gentiopicrin and swertia bitter glycoside in different herb pairs’ decoction based on gentiana were determined by High Performance Liquid Chromatograph and characteristics spectrum about gentiana was analyzed in this work.The relationship between different herb pairs and wind musculoskeletal disease and syndrome was analyzed comprehensively.Results: 1.Compared with collagen and collagen Ⅱ Ⅱcombined with wind cold dampness group, arthritis index scores of all of therapy group had an obvious decrease,there was a statistical significance(P<0.01),among them,herb pairs gentiana-clematis group had a lowest score;Compared with herb pairs gentiana-clematis group,arthritis index scores of single herb gentiana,herb pairs gentiana-mistletoe group had an obvious rise,there was a statistical significance(P<0.05).2.In the determination of 18 kinds of routine blood indexes, eight of 18 indexes in the normal group and collagen Ⅱcombined with wind cold dampness group, difference of which was statistically significant, that is to say, compared with the normal group, RBC, hemoglobin, red blood cells deposited, average red blood cell volume, average red blood cell hemoglobin, platelet deposited, mean platelet volume and platelet distribution width variation coefficient in collagen Ⅱcombined with wind cold dampness group were significantly lower(P < 0.01); Compared with the normal group, white blood cells of single herb gentian group, herb pairs gentian-clematis group, gentian-mistletoe group and gentian-tetrandra group were increased,especially in single herb gentian group,which was obviously higher, the difference was statistically significant(P < 0.01), white blood cells of herb pairs gentian-mistletoe group and gentian-tetrandra group were all increased(P< 0.05), herb pairs gentian- clematis group also had increased,but there was no statistically significant difference(P > 0.05), while collagen, white blood cells of collagen combined with wind Ⅱ Ⅱcold dampness group were decreased, there was no statistically significant difference(P > 0.05). 3.Compared with the control group,erythrocyte sedimentation rate(ESR) measured after 12 hours in gentiana group,gentiana-clematis group and gentiana-tetrandra group had decreased at a degree,there was a statistical significance(P<0.05),at the same time, ESR of gentiana group and gentiana-mistletoe group had an obvirous decrease(P<0.01).Compared with the pure collagenⅡgroup,ESR of gentiana group and gentiana-clematis group had an obvirous decrease,there was a statistical significance(P<0.01) as well as gentiana-terandra group which had a significant decrease(P<0.05).4.Compared with single herb gentiana,the content of index components including gentiopocroside in herb pairs gentiana-clematis had a bitter decrease,howver, swertiamarin in herb pairs gentiana-clematis had increased at a small degree as well as herb pairs gentiana-mistletoe,but the content of target componets called gentiopocroside in herb pairs gentiana-tetrandra had an obvious decrease,another index component called swertiamarin whose content kept an unchanged.Conclusion: In wind musculoskeletal RA rat model,compared with the model group, the efficacy of herb pair called gentiana-clematis was best,followed by gentiana-mistletoe, single herb gentiana,gentiana-terandra. To a certain extent,the peak area of indicative chemical composition called gentiopicrin and swertia bitter glycoside in different herb pairs’ decoction based on gentiana were changed.Compared with single herb gentiana,the compatibility of different herb pairs based on gentiana had devoted to an increase of total contents of substances representing all characteristic peak about gentiana.That is to say, compatibility can enhance the efficacy,however,the relevance on characteristic spectrum and clinical efficacy needs further research.
