Serum MiRNA-25 Expression In Paitients With Lung Cancer And The Clinical Significance

Objective:Lung cancer is the highest in the mortality of the malignancy tumors all the word.The morbidity and mortality are upswing year by year.It is a serius threat to human survival and health.However,the vast majority of patients have been developed to locally advanced or metastatic at the time of initial treatment.So the early diagnosis of lung cancer is the key to prolong the survival of patients.The pathogenesis of lung cancer is not clear. In recent years, along with the deep study of miRNA, we have found that miRNA may adjust the development of lung cancer in the level of post-transcriptional,Early studies have suggested that miRNA-25 is closely related to proliferation,apoptosis, drug resistance in lung cancer. In our trial, throught studying the serum miRNA-25 expression in lung cancer patients and healthy human,we discuss differential expression of serum miRNA-25 in different pathological types of lung cancer, and further analysis the clinical diagnostic value of the serum miRNA25 in patients with lung cancer and tassess the relationship between the serum miRNA25 and clinical factors.Methods:Eighty-eight serum sampals from 30 non-small cell lung cancer,28 small cell lung cancer and 30 healthy controls were collected as research objects.Serum levels of miR-25 were detected by quantitative real-time PCR analysis.Then the clinical value in the diagnosis of lung cancer was evaluated using ROC curves by calculating sensibility and specificity.Results:The serum level of miRNA-25 was significantly higer in non-small cell lung cancer group(P=0.001)and small cell lung cancer group(P=0.000)than that of healthy control group;the serum level of miRNA-25 in SCLC was higher than NSCLC,and the differential expression was statistically significant(P=0.001);Spearman rank correlation indicate there was significantly correlation between serum miRNA-25 and the pathological type(r = 0.422, P = 0.001), tumor stage(r = 0.370, P = 0.004), lymph node metastasis(r = 0.670, P = 0.000), tumor markers(r = 0.372, P = 0.004).Then there was nothing to do with age,sex,the history of smoking,all the value of P were bigger than0.05.The area under the ROC curve of NSCLC was 0.659 when 1.854 was sellected as the critical value,the sensitivity and specificity were 56.7% and 90% respectively.While the ROC plot derived from SCLC yielded an area under the curve(AUC) of 0.869,the critical value was 2.424,and the sensitivity and specificity were 78.6% and 97%respectively.Conclusion:The serum level of miRNA-25 is significantly elevated in lung cancer patients,expesially in SCLC,it may be used as a potential biomarker in diagnosis of lung cancer.
