Objective:To observe the effect of NAC on apoptosis and oxidative stress in cardio-renal syndrome rats.Methods:20 healthy adult male rats were randomly divided into 4 groups(sham, sham NAC, AAC, AAC NAC), then we did abdominal aortic ligation(AAL) to each rat of AAC group. The right time after surgery, we gave the NAC to sham NAC group and AAC NAC group as pharmacological intervention for 4 weeks. Also we gave saline as placebo to each one of sham and sham AAC group. After that we detected the heart function of rats by echocardiography and BUN、SCr、MDA by biochemical Kit. Also we used western blot and immuno-histochemistry to test the expression of Bax、Bcl-2、Caspase-3 and Caspase-9 in rat kidney tissue. finally, we used HE staining and TUNEL to explore renal pathological changes and apoptosis of kidney tissue.Results:The result of echocardiography showed that the AAC group had left ventricular hypertrophy and poor cardiac function than sham group. HE staining showed renal tubular necrosis and glomerulus pyknosis in AAC group. Nevertheless, NAC AAC group significantly reduced left ventricular hypertrophy and kidney pathology damage compare with AAC. Compare with sham group,AAC group showed that BUN,SCr,MDA and apoptosis related protein Bax increased obviously, anti-apoptosis relatedprotein Bcl-2 decreased, Caspase-9 and Caspase-3 significantly increased. Compare with AAC,AAC NAC group has showed that Bcl-2 obviously increased. BUN,SCr,MDA,Bax, Caspase-9, Caspase-3 are significantly decreased. In TUNEL experiment,we can also found the same results. There is no difference between Sham group and sham NAC group above all.Conclusion:NAC has obvious protective effect on renal function of cardio-renal syndrome.The mechanism may be associated with inhibition of mitochondrial pathways of apoptosis and oxidative stress.
