Objective To study the relationship between ACTN1 and renal cell carcinoma. Recently, studies have af?rmed that ACTN1 is associated with several human cancers, including lymphoma, laryngeal squamous cell carcinoma, prostate cancer, and liver cancer, which suggests ACTN1 as a potential therapeutic target in cancer. However, the functional role of ACTN1 in human renal cancer remains unclear.Methods Herein, we detected the expression of ACTN1 in 38 patients with renal cancer at them RNA and protein levels. Moreover, we employed lentivirus-mediated small inter RNA(siRNA) to knock down ACTN1 expression in two human renal cell carcinoma cell lines, 786-0 and A704. We performed sequencing analysis to explore the influence factors of ACTN1 over-expression in RCC.Results We found that the expression of ACTN1 was speci?cally upregulated in cancer tissues at them RNA and protein levels compared with the adj acent normal tissue. Functional analysis indicated that ACTN1 silencing signi?cantly inhibited renal cell carcinoma cell proliferation and cell cycle progression and induced apoptosis. Our ?ndings suggest that knockdown of ACTN1 by siRNA may be a potential therapeutic approach for the treatment of renal cancer.Conclusions 1. ACTN1 plays an important role in the processing of tumorigenesis and development of human RCC, ACTN1 may be as one of oncogenes in RCC. 2. Knockdown of ACTN1 by siRNA could inhibit the growth, invasion and motility of RCC cell-lines, which indicate that ACTN1 influence the proliferation, aggressive and metastatic of RCC tumor. 3. Down-regulation of ACTN1 could induce apoptosis and block cell cycle of RCC cells.
