| With the increasing demand of Chinese herbal medicine in medical market both at China and abroad, the shortage of traditional production mode appeared gradually in Traditional Chinese Medicinal Materials quality, production efficiency and the rate of input-output, which seriously restricted the development process of modern industrialized production of modern Traditional Chinese Medicine.In this paper, we take Wendang as the example, to develop the optimized original processing techniques of fresh Wendang based on studying the influences of drying, vacuum freeze drying,microwave drying and traditional processing method on the active ingredients and antioxidant activity of Wendang herbs. Firstly, dual wavelength High Performance Liquid Chromatography(HPLC) fingerprints of 18 WPP herbs samples prepared under 4 different drying methods were developed. The change of 5-hydroxymethyl furfural(5-HMF) and other major chemical components including polysaccharid, total flavonoids, alcoholic extracts, aqueous extracts,lobetyolin and atractylenolide III in the 18 Wendang pieces was examined, and the DPPH free radical scavenging activity of all samples was studied. Then, the correlation of HPLC fingerprint and antioxidant activity was investigated by partial least squares(PLS), and the correlation peaks were identified by HPLC-MS/MS technique. The results showed that seventeen common peaks(P1-P17) were labeled in the chromatographic fingerprints at wavelength of 267 nm, nine common peaks were labeled in the chromatographic fingerprints at wavelength of 320 nm(P1*-P9*), the area of the twenty-six common peaks accounted for 90% of total peak areas. The ratio of common peaks area of fingerprint / sample weight(g) of piece S4 in 18 Wendang pieces sample over traditional processing was highest(82.49%). The spectrum-efficacy relationship results indicated that 9 characteristic peaks was positively correlated with DPPH clearance rate, P13 which had the greatest contribution to antioxidant activity was identified as lobetyolin by using HPLC-MS/MS as well as comparing with the standard.Taking into comprehensive consideration, the optimized the processing and concocting integration technologies of Wendang pieces was that fresh WPP was baked to water content of50% at 80 °C and took out, then Cut 0.3-0.5cm thick film and dried to constant weight at 50 ℃in the oven. The process could keep the contents of lobetyolin, polysaccharid, atractylenolide III,alcoholic extracts and aqueous extracts in Wendang medicinal materials at largest extent.Meanwhile, the prepared Wendang pieces had the stronger antioxidant activity. This paper was also benefit for the effective utilization of origins and the development of local economy. |
