| OBJECTIVE The aim of this study was to investigate the protective effects of Si Wei La Gen Cai water extract on myocardium against ischemia reperfusion injury in rats.METHODS 48 SPF wistar rats were randomized into sham operated group(group SO), ischemia reperfusion injury group(group I/R), salviae miltiorrhizae(1.75 g·kg-1·d-1) group as positive control,and low(0.7 g·kg-1·d-1),middle(1.4 g·kg-1·d-1) and high(2.1 g·kg-1·d-1) dose group of Si Wei La Gen Cai water extractr Cochlearia Officinalis. After 10 days’ intragastric administration,group I/R underwent ligations of anterior descending branch of left coronary artery,receiving infusion of 90 minutes immediately after 30 minute period of ischemia.No ligation was applied in group SO.We first observed electrocardiogram(ECG) data, and then performed blood perfusion. Blood was collected at the end of abdominal aorta in order to detect the activities of creatine phosphokinase(CK), lactate dehydrogenase(LDH), superoxide dismutase(SOD), glutathione peroxidase(GSH-PX) in serum, and the contents of malondialdehyde(MDA) and troponin I(c Tn I). At last,we observed left ventricular geometry with transmission electron microscope(TEM).RESULTS CK and LDH release in serum were decreased in Si Wei La Gen Cai water extract group comparative to group(I/R)(P<0.05).SOD and GSH-PX activity(P<0.05)were found significantly high, whereas MDA(P<0.05)and c Tn1(P<0.05) contents were reduced. Moreover, in both high and middle dose of the Si Wei La Gen Cai water extract groups, changes in mitochondria geometry, which is the ultra structure of myocardial I/R injury region, can be alleviated.CONCLUSION These results indicated that both middle and high doses of Si Wei La Gen Cai water extract have obvious protective effects on I/R injury in myocardium in rats. |
