Objective To investigate the expression of Gli2 and TGF-β1 in non-small cell lung cancer(NSCLC) and their clinical significance and prognosis of non-small cell lung cancer.Methods Eighty-nine NSCLC specimens and paraneoplastic normal lung tissue from conventional surgery were confirmed by histology.all cases had not been treated with radiotherapy and chemotherapy before their surgeries.Expression of Gli2 and TGF-β1 were detected by Immunohistochemistry on paraffin sections.According to the patients’ survival status from follow-ups, we could study the relationship between expression of Gli2、TGF-β1 and prognosis. 25 cases had been collected from fresh specimens of non-small cell lung cancer in 2014, We could use RT-PCR to detect the expression of Gli2 m RNA in non-small cell lung cancer.Result 1. In non-small cell lung cancer the positive rates of Gli2,TGF-β1expression(respectively94.4%and100%) were higher than the positive rates(13.5%and30.3%) of those in adjacent normal lung tissue, and there was statistical significance(both P <0.01).The rates of high expression of Gli2,TGF-β1were 73.0% and 78.7%respectively, The high expression of Gli2 and TGF-β1 was positively correlated in non-small cell lung cancer(r = 0.438, P <0.01). 2. There were significant differences in the expression of Gli2 between different TNM stages, hematogenous metastasis and lymph node metastasis in NSCLC group. There were significant differences in the expression of TGF-β1 between different tumor size, hematogenous metastasis and lymph node metastasis in NSCLC group. The expression of Gli2 was positively correlated with TGF-β1(r=0.438, P <0.01). 3. According to the patients’ survival status from follow-ups, we could get the result that the three years survival rate was 83.3 % in the group of Gli2 low expression and was 58.5% in the group of Gli2 high expression, the difference was statistically significant(Log-rank c2=5.246 P <0.05)。The three years survival rate was 77.8 % in the group of TGF-β1 low expression and was49.2% in the group of TGF-β1 high expression, the difference was statistically significant(Log-rank c2=4.761 P <0.05)。 4. RT-PCR results showed that expression of Gli2 m RNA(0.82±0.13) in lymph node metastasis samples than expression of Gli2 m RNA expression(0.54±0.03) in those samples without lymph node metastasis, and this difference was statistically significance(t =2.334 P <0.05).The expression of m RNA(0.89±0.13) in the group of stage Ⅲ was higher than those(0.51±0.03)in the group of stage Ⅰ and Ⅱ, and this difference was statistically significant(t=3.458 P <0.01)。Conclusion 1. The expression of Gli2, TGF-β1 in non-small cell lung cancer was higher than those in normal lung tissue, and the expression of Gli2 and the expression of TGF-β1 in non-small cell lung carcinoma were positively correlated. 2. There were significant differences in the expression of Gli2 between different TNM stages, hematogenous metastasis and lymph node metastasis in NSCLC group. The higher TNM stage was, the expression of Gli2 was higher, that expression of Gli2 in lymph node metastasis samples than expression of Gli2 in those samples without lymph node metastasis. The expression of Gli2 in vessel metastasis samples was higher than expression of Gli2 in those samples without vessel metastasis. There were significant differences in the expression of TGF-β1 between different tumor size, hematogenous metastasis and lymph node metastasis in NSCLC group. with tumor size larger, the expression of TGF-β1 was higher, that expression of TGF-β1 in lymph node metastasis samples than expression of TGF-β1in those samples without lymph node metastasis. The expression of TGF-β1 in vessel metastasis samples was higher than expression of TGF-β1in those samples without vessel metastasis. The expression of Gli2 in gene level was associated with TNM stage, lymph node metastasis. 3. The high expression of Gli2、TGF-β1 were associated with with poor prognosis and it suggested that they could be used as an indicator of prognosis in non-small cell lung cancer。...
