ObjectiveIt to study the immune regulation mechanism of Forsythoside on sepsis rats induced by cecal ligation and perforation(CLP).ContentsWe detect survival rate and the influence of the temperature change in each group. ELASA was used to detect changes of serum complement(C5, C5a) in serum and cytokines(TNF-α, IL – 1β, IL – 6). Western blotting was used to test changes of CD88 protein expression of lung, liver and kidney.Methods210 male wistar rats were randomly divided into control group(n=15),sham-operation group(n=15) and model group(n=180). The model group was randomly divided into CLP model group and Forsythoside group(each subgroup n=90).The latter two model group according to different time points(24 h, 48 h and72 h) were randomly divided into 3 groups(each subgroup n=30). The rats of model group received cecal ligature and puncture(CLP) operation. Forsythoside group respectively at 0 h, 12 h, 24 h, 48 h, 60 h were given Forsythoside acetate injection200μg. Control group, sham-operation group, CLP model group were given volume of stabilizer. The content of changes of serum complement(C5, C5a) in serum and cytokines(TNF-α, IL-1β, IL-6) were measured with ELASA and changes of CD88 protein expression of lung, liver and kidney were measured with Western blotting.Results1 The numbers of Forsythoside group in each observation point are much less than control group, and activity, reaction and eating were obviously better than control condition.2 The survival of Forsythoside group was 80.00%, 63.33%, 36.67%respectively at 24 h, 48 h and 72 h. The survival of control group was 63.33%,53.33%, 26.67% respectively at 24 h, 48 h and 72 h.3 Forsythoside can improve the low temperature after CLP at 24 h and 48 h. In comparison with Forsythoside group compared and control group at 48 h, thisdifference had statistical significance P=0.005).4 The anatomy of the visible: Normal group and control group had no obvious abdominal cavity, swelling and adhesion. We can find intra-abdominal exudate is more, the stench, diffuse abdominal cavity infection and intestinal edema, liver and kidney serious congestion in CLP group.5 Serum complement C5: In comparison with Forsythoside group compared with control group at 24 h and 48 h, this difference had statistical significance(P=0.001). Complement C5a: In comparison with Forsythoside group compared with control group at 24 h, this difference had statistical significance(P= 0.000). TNF-α:In comparison with Forsythoside group compared with control groupat 24 h, this difference had statistical significance(P=0.017). IL-1β: In comparison with Forsythoside group compared with control group at 24 h, this difference has statistical significance(P= 0.000). IL-6: In comparison with Forsythoside group compared with control group, this difference had no statistical significance at the same time.6 Forsythoside can reduce the expression of lung tissue CD88 expression at 72h(P=0.036), the expression of liver tissue CD88 expression at 24 h and 72 h(P=0.010,P=0.000). Forsythoside can mild inhibite the expression of kidney tissue CD88 expression.Conclusion1 Forsythoside can improve the survival rate in rats by cecal ligation and perforation(CLP) at 24 h, 48 h and 72 h. Forsythoside can improve the low temperature after CLP at 24 h and 48 h.2 Forsythoside can reduce the level of serum complement C5 and C5 a at 24 h.Forsythoside can reduce the level of serum TNF-α at 24 h and 48 h. Forsythoside can reduce the level of serum IL-1β at 24 h. Forsythoside can mild inhibite the expression of serum IL- 6.3 Forsythoside can reduce the expression of lung tissue CD88 expression at 72 h. Forsythoside can reduce the expression of liver tissue CD88 expression at 24 h and72 h. Forsythoside can mild inhibite the expression of kidney tissue CD88 expression.
