Objective we investigated the immune mechanism of Forsythoside`s effects on sepsis rats induced by cecal ligation and perforation(CLP).It could provide immunological evidence of targeted therapy of Forsythoside.Methods Established sepsis models in rats by cecal ligation and perforation(CLP).210 male wistar rats were randomly divided into control group(n=15),sham-operation group(n =15) and model group(n=90) and treatment group(n=90).model group and treatment group according to different time points(24h, 48 h and72h) were randomly divided into 3 groups(each subgroup n=30).The CLP were operated in the rats of model group and treatment group.Treatment group were respectively given Forsythoside 0.5 ml by acetate injection at 0h,12 h,24h,48 h and60h.Control group,sham-operation group and model group were all given the isometric normal saline in the same way. We messured the changes of serum cytokines(TNF-α,IL – 1β,IL – 6,MIF and HMGB1) by means of ELISA and the changes of CD88 m RNA expression of lung, liver and kidney tissue by means of q PCR.The histologic changes of lung tissue,liver tissue and kidney tissue of sepsis rats were observed by means of HE staining.We also observed the expression of CD88 m RNA on the cell membrane of lung,liver and kidney tissue by means of Immunohistochemistry(IHC).Results1 Serum TNF-α: The level of treatment group was significantly lower than model control group at 24h(P=0.017). Serum IL-1β: The level of treatment group was significantly lower than model control group at 24h(P=0.000). Serum IL-6: The level of treatment group had no significantly difference with control group. Serum MIF:The level of treatment group had no significantly difference with control group.Serum HMGB1: The level of treatment group had no significantly difference with control group.2 In the lung tissue part,in comparison with treatment group and control group at72h、48h,the difference of the expression of CD88 m RNA on the membrane detected by q PCR both had statistical significance(P=0.005,P=0.000). there were no statistical differences in other groups compared at the same time.3 Through the HE staining,there were no significant histologic changes of lung tissue,liver tissue and kidney tissue of sepsis rats such as oedema or necrosis.4 There was a high expression of C5 a receptor detected in the membrane of by IHC in the cell membrane of lung tissue while a low expression of C5 a receptor detected by IHC in the liver and kidney tissue in the sepsis rats.Conclusion1 Forsythoside can reduce the level of serum TNF-α at 24 h and 48 h. Forsythoside can reduce the level of serum IL-1β at 24 h.2 Forsythoside can reduce the expression of lung tissue CD88 m RNA at 72 h and 48 h.
