Background: Chemical liver injury is caused by chemical substances that have hepatotoxic effects. With the development of economy and the improvement of people’s living standard, the incidence of chemical liver injury has increased year by year, which endangered people’s health seriously. Since there have not yet formed a national brand of liver health food, the market for liver health food especially using Chinese herbal medicine as raw materials is expanding.Obejctive: Our aim is to develop a new liver health food in kind of capsules by using Ginseng, ganoderma lucidum extract, gynostemma extract, rhodiola extract, schisandra fruit extract as raw materials, which have auxiliary protective effects on liver injury. Toxicology test, security and toxicology tests, function test, and quality standards establishment were performed to estimate the dosage and demonstrate the liver protective effects. The aim of this product was to have auxiliary protective effects on people with chemical liver injury.Methods: 1. Based on a large number of literature, and combining traditional chinese and western medical theories, ginseng, ganoderma lucidum extract, gynostemma extract, rhodiola extract, schisandra fruit extract were used as raw materials. And the optimal production process were screened by experiments. The dosage form was decided to becapsule, and the health food was named liver-protecting capsules. 2. SPF kunming graded mice and Wistar rats were used to do security and toxicology tests, which including acute oral toxicity test, genetic toxicity test safety toxicology test and 30 days feeding test, and the acute toxicity reaction, death, etc. of mice were observed. 3. The model of alcohol induced liver injury in mice was set up. After liver-protecting capsules were used to feed mice, the MDA, GSH and TG contents in liver homogenate were measured and liver tissue pathology analysis were performed.Result: 1. The formula of liver-protecting capsules were determined, the technological process of production were opimized, and the quality standard were established. 2. Security and toxicology tests showed that MTD value was greater than 20.0g/kg·bw. The results of two mutagenic tests including mice bone marrow cell micronucleus test and sperm malformation test were negative, and it was not found obviously toxic effects in 30 days feeding test. 3. The function tests showed that the liver-protecting capsules had auxiliary protective effects on alcoholic liver injury in mice.Conclusion: On the basis of decision criteria in the health food inspection and evaluate technical specification, the product was nontoxic and it had auxiliary protection on alcoholic liver injury in mice.
