ObjectiveTo study the inhibitory effect of Human embryonic stem cells on the HepG2 cells in vitro.To investigate the inhibitory effect of supernatant from co-culture of Human embryonic stem cells with tumor MDA-MB-231 cells on the MDA-MB-231 cells of breast cancer in vitro.Methods1. The co-culture system of Human embryonic stem cells(H9) and liver cancer HepG2 cells was established.The effect of supernatant on the biological behavior of tumor cells was observed by microscope, the flow cytometry was used to detect the apoptosis of tumor cells and the cell cycle alteration.2. The direct co-culture system of Human embryonic stem cells(H9) and breast cancer MDA-MB-231 cells was established, and the supernatant from co-culture cells was collected to treat MDA-MB-231 cells. The effect of supernatant on the biological behavior of tumor cells was observed by microscope. MTT assay was used to detect the cell proliferation. Hoechst staining and flow cytometry were used to detect the apoptosis of tumor cells. Transwell assay was used to detect the migration and invasion of tumor cells.Results1.in the process of co-culture, the growth of hepatoma cells was inhibited. With the extension of the culture time,cells decreased gradually, and occurred. Signs of aging or apoptosis. Flow cytometry test results showed that the apoptosis rate of hepatoma cells was significantly increased, and the cell cycle was blocked in the G0/G1 phase.2.After treating with the supernatant from co-culture of Human embryonic stem cells with tumor MDA-MB-231 cells, there were cell morphological changes and signs of apoptosis on MDA-MB-231 cells. Typical apoptotic morphological changes such as nuclear shrinking,and nuclear fragmentation could be observed under fluorescent microscope after staining with Hoechst 33258.Cell apoptosis rate increased; cell survival rate and cell invasion, migration were decreased. While the control group of MDA-MB-231 cells were almost not affected.ConclutionsThe human embryonic stem cells had an inhibitory effect on HepG2 cells in vitro. The supernatant of co-culture cells had some anti-tumor effects on MDA-MB-231 cells in vitro. Therefore,the human embryonic stem cells had a certain anti-tumor effect in vitro.
