Objective: To evaluate the expression of IL-37 in patients with allergic rhinitis and its regulation on the level of cytokine IL-1β、IL-6、IL-27、IL-10 from PBMCs. And futher explore the effects of rIL-37 on the expression of cytokine IL-17 and IL-10 from CD4~+ T cells.Methods: Thirty-nine patients with allergic rhinitis and 43 healthy controls were enrolled in this study.Peripheral blood samples from all patients and healthy controls were obtained. The cell proportion and expression of IL-37 from PBMCs were measured by FCM、ELISA and RT-PCR. The level of cytokine IL-1β、IL-6、IL-27、IL-10 in the culture supernatants of PBMCs were detected by ELISA and RT-PCR from PBMCs with the stimulation of rIL-37. We analyze the effects of rIL-37 on the expression of cytokine IL-17 and IL-10 from CD4~+ T cells.Results: Our results showed that the expression of IL-37 from PBMC was significantly decreased in allergic rhinitis patients as compared with normal controls(p<0.05). Moreover, in allergic rhinitis patients, we found obviously decreased levels of IL-1β and IL-6 and increased level of IL-27 in rIL-37-stimulated PBMCs culture supernatants. rIL-37 has no effect on the production of IL-10 by PBMCs. Finally, we showed that IL-37 reduced IL-17 production, IL-37 has no influence on the production of IL-10 by CD4 +T cells.Conlusion: Our study demonstrated that expression of IL-37 from PBMC was significantly decreased in allergic rhinitis patients. Furthermore,rIL-37 acts on PBMCs and CD4~+T cells to inhibit the inflammatory response of allergic rhinitis.
