Objective: By building experimental autoimmune prostatitis model to observe the influence of curcumin on the cytokines,we explore its possible mechanisms in chronic prostatitis/chronic pelvic pain syndrome.Methods: Sixty healthy male Sprague Dawley rats were randomly and equally split into four groups,names the normal group,the model group,the intraperitoneal curcumin group and the positive control group.The experimental autoimmune prostatitis rats model were made by multiple hypodermic injection of prostate extract mixed with adjuvants.After the autoimmune prostatitis model was built,every group was treated for four weeks.The levels of TNF-α in prostate tissues of the rats were detected by ELISA assay. The expressions of NF-κ B and COX-2 in rat Prostate tissue were respectively measured by immunohistochemistry, immunofluorescent and western blot.Results:The levels of IL-1β,IL-10,TNF-α,NF-κ B and COX-2 in prostate tissues were significantly lower in the intraperitoneal curcumin group and positive control group,by contrast with the model groups(P<0.01).Treatment with curcumin(100 mg/kg) reduced the levels of IL-1β,IL-10,TNF-α to the positive control group(P<0.01).But the expression of COX-2 and NF-κ B had no great difference between the positive control group and the intraperitoneal curcumin group(P>0.05).Conclusion: Curcumin is efficient for experimental autoimmune prostatitis rat model, and the mechanism may be associated with the reduction on the cytokines TNF-α,NF-κ B and COX-2 in prostate tissues.
