Objective To explore the apoptosis and the expression of SIRT1 in human degenerative cartilage endplate(CEP) and to investigate whether SIRT1 could suppress the apoptosis of the degenerative endplate chondrocytes. Methods CEP tissues were obtained from the lumbar spine surgery patients with lumbar disc herniation(LDH)and lumbar vertebrae fracture(LVF) of the First Affiliated Hospital of Chongqing Medical University(Chongqing, China) from June 2014 to May 2015,the degenerative levels of the intervertebral disc was assessed by Pifrrmann grades.The cell morphology and number of the CEP were detected by HE staining.The expression of the SIRT1,caspase3, collagenⅡwere detected by Immunohistochemistry. Endplate chondrocytes were cultured and the second generation CEP cells were used to finish the experiment.The expression of the SIRT1,Bcl2 and caspase3 of the CEP cells were detected by westernblotting after treated with resveratrol and SIRT1-siRNA or nicotinamide,the apoptotic incidence was measured by flowcytometry. Results The cell types were chondrocytes and the number of cells were decreased in the degenerative CEP tissues。The expression of the SIRT1 and collagenⅡ were decreased in the degenerative CEP tissues and the apoptosis related protein caspase3 was increased in the degenerative CEP tissues. The protein expression of the SIRT1 and Bcl2 were increased and the expression of the caspase3 was decreased in the degenerative CEP cells after treated with resveratrol, the apoptotic incidence was also decreased. The protein expression of the SIRT1 and bcl2 were decreased and the expression of the caspase3 was increased in the degenerative CEP cells after treated with SIRT1-siRNA or nicotinamide, the apoptotic incidence was also increased. Conclusion Apoptosis may play an important role in the human CEP degeneration and the cell apoptosis increase in the degenerative CEP tissues,and SIRT1 could suppresses the apoptosis of the degenerative CEP cells.
