Purpose:The Glioblastoma, for short glioma, is one of the mostcommon malignant tumor in the central nervous system of human beings.Itmakes the clinical treatment difficult because of its invasive growth, which isthe main cause of leading to death.Our country is one of the country which hasthe high incidence of gliomas.The number of the infected and the death due tothe gliomas is in the forefront of which is caused by various malignanttumors.It is a world wide problem in the treatment because of its invasivegrowth.The occurrence and development is the result of comprehensiveinfluence by multi-factor and multi-gene.Glioma is the primary tumor in thehead,and many apoptotic genes,proto-oncogenes,tumor suppressor genes arerelated with the occurrence of glioma.Those genes acts in different segment oftumor cell proliferation and apoptosis,such as apoptosis inhibition orimbalance,can lead to the excessive proliferation of tumor, deciding thegrowth,metastasis and prognosis of tumor.With the development of molecularbiology research,the mechanism, which explains that the many cell adhesionmolecules and hydrolyzing protease involve in the occurrence, development,invasion and metastasis of tumor, is elucidated.But the reports about theglioma are few.So it is necessary that we study the mechanism of development, growth and invasion of gliomas from molecular level.Cell apoptosis is a process of a series of waterfall initiative death fromthe simulation by various death signal,which is regulated by variousgenes.And this is closely related to the caspase3family and bc12family.Theinteraction of apoptosis, proliferation and anti-apoptotic in tumor celldetermines the development degree of tumor biological behavior.At present,we study more on the behavior of caspase3and bc12in the gastrointestinalcancer(gastric cancer, liver cancer, colon cancer, pancreatic cancer and so on) and genitourinary tumors(breast cancer, uterine cancer, ovarian cancer, renalcancer, bladder cancer)and so on.But the research on them expression inhuman glioma is relatively few.The glioma is a cerebral tumor,in-depth studyon the behavior of caspase3and bcl2in the glioma and their relationship withclinical pathological type of glioma,which may become a new approach forthe diagnosis, prognosis and treatment of glioma.Our subject mainly study thesignificance on caspase3and bcl2in occurrence, development and prognosticof glioma by detecting the expression of caspase3and bcl2in gliomas ofdifferent levels.Methods:We have collected60cases of human glioma which aresurgically removed in department of neurosurgery, at second hospital of hebeimedical university from December2012to April2013.By the world healthorganization (WHO) classification of standards, the Ⅰ and Ⅱ brain gliomacells were divided to low level group, the Ⅲ and Ⅳ brain gliomas weredivided to high level group.23cases of brain gliomas in low level group and37cases of brain gliomas in high level group.Those cases have completemedical records and have been confirmed by the department ofpathology.Another11cases of normal brain tissue which is reduced pressurein the brain due to trauma or cerebral hemorrhage are made to contrast. Thepatients are not carried out any anti-cancer therapy(chemotherapy)before thesurgery. All samples are saved immediately after cutting the tissue in thesurgery, and they are fixed by10%formalin and saved in the liquid nitrogen.In human glioma, expression of bc12and casepase3in protein level andmRNA level were detected by using immune histochemical (IHC) andRT-PCR method.Results:1The result of IHC shows:The expression positive rate of bcl2in normalbrain tissue group, low–grade glioma group and high-grade glioma group arerespectively9%,78%,100%.And the caspase3expression positive rate are0%,100%,84%.The expression positive rate of bcl2and caspase3in the threegroups have a significant statistical difference(P=0.0000).And the pairwise comparison between groups is a significant statistical differences(P<0.05).With the increased degree of tumor, the level of bcl2is increased and the levelof caspase3is decreased.The expression of bcl2and caspase4in the threegroups is negatively correlated(r=-0.814,P=0.000).But there is no correlationin other groups,this may be caused by less samples.2The result of RT-PCR shows:the expression of bcl2and caspase3in thethree groups(normal brain tissue, low–grade glioma and high-grade glioma)have a significant statistical difference (P<0.05).And the pairwisecomparison between groups is a significant statistical differences(P<0.05).With the increased degree of tumor, the level of bcl2is increased and the levelof caspase3is decreased.And the expression of bcl2and caspase3in the threegroups is negatively correlated(r=-0.254,P=0.032).And the expression ofthem is also negatively correlated in the tumor groups(low–grade glioma andhigh-grade glioma).But it is no correlation in other groups following thechecked level of P <0.05.Conclusions:1The expression of bcl2in the three groups(normal brain tissue, low–grade glioma and high-grade gliomas) and the pair groups has a significantstatistical difference.With the increased degree of tumor, the level of bcl2isincreased and is highest in the high-grade glioma. The expression of bcl2isassociated with malignant degree of gliomas.2The expression of caspase3in the three groups(normal brain tissue, low–grade glioma and high-grade gliomas) and the pair groups has a significantstatistical difference.With the increased degree of tumor, the level of bcl2isdecreased and is highest in the low-grade glioma. The expression of caspase3is associated with malignant degree of gliomas.3Down regulation of Caspase3expression be closely related withmalignant progression of gliomas. Bcl2plays an important role in theoccurrence and development of glioma. Caspase3and Bcl2may be a potentialprognostic indicators of glioma.4Caspase3and Bcl2expression in gliomas and have a significant correlation that with the Caspase3expression reduced, Bcl2expressionincreased.
