Objectives:To explore the effects of pcDNA3.1-PLCD1 plasmid on the expression of PLCD1 mRNA,PLCD1 protein and the proliferation,cell cycle,apoptosis,invasion, migration of human pancreatic cancer cells CAPAN-1 and BXPC-3.Methods:First,we cultured normal human pancreatic ductal epithelial cells HPDE6C7 and pancreatic cancer cell lines CAPAN-1, PANC-1, ASPC-1 and BXPC-3,and extracted RNA then conducted reverse transcription and semi-quantitative RT-PCR to screen the lower expression of PLCD1 in pancreatic cancer cell lines.The above selected pancreatic cancer cell lines CAPAN-1 and BXPC-3 were transfected with plasmid mediated by liposome,and it have pcDNA3.1-PLCD1 and pcDNA3.1 transfection group.To detect the expression of PLCD1 mRNA by RT-PCR and PLCD1 protein by western blot on transfected cells,The proliferation of transfected pancreatic cancer cells were detected by CCK-8 and colony formation assays, the apoptosis and cell cycle of transfected pancreatic cancer cells CAPAN-1 and BXPC-3 were examined and analysised by flow cytometry,transwell assay detected the invasion and migration abilities of the above transfected pancreatic cancer cells CAPAN-1 and BXPC-3.Results:The pancreatic cancer cell lines CAPAN-1 and BXPC-3 which have lower expression of PLCD1 mRNA were selected triumphantly and used in follow-up experiments.The pcDNA3.1-PLCD1 transfection group has a high expression of PLCD1 mRNA and PLCD1 protein,and also can restrain proliferation,invasion and migration of CAPAN-1 and BXPC-3 cells.Meanwhile it can promote cell cycle was arrested in G1 phase and cell apoptosis.Conclusion:PLCD1 gene can promote cell apoptosis and cell cycle was arrested in G1 phase, and inhibit cell proliferation, migration and invasion of pancreatic cancer cell lines CAPAN-1 and BXPC-3.
