Objective To investigate the influence of different doses of Vitamin C(ascorbic acid)on Antitumor Effect of Oxaliplatin on a nude mouse model of human gastric carcinoma. Methods A model of subcutaneous transplanted with human gastric carcinoma cells(SGC-7901) was created in nude mice, which were divided into eight groups according to random number table method, and each group had 10 mice. Group A was given 5%Glucose; group B was given Oxaliplatin 6 mg/ kg; group C、D、E were respectively given low, medium, and high doses of Vitamin C 100、200、400 mg/ kg; group F、G、H were respectively given low, medium, and high doses of Vitamin C 100、200、400 mg/ kg and Oxaliplatin 6 mg/ kg. Glucose and Vitamin C were given through an intraperitoneal injection once a day, Oxaliplatin was given through an intraperitoneal injection every three days, and continued for 15 days. Observing the life state of the nude, detecting the weight of the mice and the volume of tumor, calculating the inhibitory rate, drawing transplanted tumor growth curve. Killing animals after drug withdrawal, blood routine, liver and kidney functions were checked, and tumor and liver tissues were examined in histology. Results The interaction consisted in Vitamin C and Oxaliplatin(p<0.01), was antagonistic, the higher concentration of Vitamin C, the stronger antagonism of antitumor effect of Oxaliplatin; group C、D、E did not inhibit tumor growth(p>0.05); group B、F、G、H could inhibit tumor growth(P<0.01), with the concentration of Vitamin C increasing, the inhibition rate was decreased. Conclusion Different concentrations of Vitamin C did not inhibit gastric cancer cells growth; Vitamin C was antagonistic on Oxaliplatin, showing dose dependent.
