Objective:This study was designed to established asthma model by intraperitoneal injection and atomized inspire and chilling by cold air, drink cold water and cold water bath to detect the change of cAMP-PKA pathway of asthma in rats treated by lingganwuweijiangxin decoction to research the effect of lingganwuweijiangxin decoction to asthma.Methods:90 SPF Wistar rats were divided into six groups randomly and equally in weight,which were as follows:blank froup,model group, lingganwuweijiangxin decoction high-dosegroup, lingganwuweijiangxin decoction middle-dosegroup, lingganwuweijiangxin decoction low-dosegroup and positive group.Rats were treated with normal saline in blank group.The Rats in othet groups were treated with ovalbumin(OVA) by intraperitoneal injection and atomized inspire and chilling by cold air, drink cold water and cold water bath were employed to establish asthma animal model.Then,rats in groups which were treated in Chinese medicine were given lingganwuweijiangxin decoction.Rats were treased with dexamwthasone,the rest rats were treasted with normal saline gabage in the same volume.All rats were treated one time per day and continued 27 days.After these steps,using pentobarbital sodium made rats narcosis,cut off arteria cruralis to get blood and splited chest to get lung tissue.Then got a small amount of lungtissue fix by paraffin embedding with HE and immunohistochemical further diagnosis,and the rest lung tissue were collected by ELISA,RTqPCR and Western blot experiments.Results:1. With the blank group, the treatment group and the untreated group light microscopy of lung tissue in varying degrees of airway damage and inflammatory cell infiltration of inflammatory changes. lingganwuweijiangxin decoction flavors after treatment compared with the model group, the treatment group rat lung tissue biopsy scores were lower,with statistical significance(P <0.05).2. Compared with the blank group, the levels of cAMP,PKA, AQP5 and CREB in asthma model group rats reduced greatly(P<0.05); compared with the model group, the levels of cAMP,PKA, AQP5 and CREB in the treatment group rats increased(P<0.05); and increased dramatically in the treatment group of high-dose.3. Compared with the blank group, the mRNA expression of cAMP,PKA, AQP5 and CREB in asthma model group rats reduced greatly(P<0.05); compared with the model group,the mRNA and the protain expression of cAMP,PKA, AQP5 and CREB in the treatment group rats increased(P<0.05).4. Compared with the blank group, the protain expression of cAMP,PKA, AQP5 and CREB in asthma model group rats reduced greatly(P<0.05); compared with the model group,the mRNA and the protain expression of cAMP,PKA, AQP5 and CREB in the treatment group rats increased(P<0.05).Conclusion:1. In this study, cold drink V asthma rat model of pulmonary rehabilitation is successful, and lingganwuweijiangxin decoction soup flavors can be adjusted by airway fluid secretion, reduce inflammation and thus play a therapeutic cold drink underlying pulmonary asthma action;2.The expression levels of cAMP, PKA, AQP5, CREB gene and protein were risingly,indicating that cold drink underlying lung asthma rat lung tissue the expression levels of cAMP, PKA, AQP5 and CREB can reflect the extent of damage lung tissue, and then infer the expression levels of cAMP, PKA, AQP5, CREB assessment of asthma and the effects of treatment decision may potentially instructive;3.Lingganwuweijiangxin decoction flavors therapeutic effect may be related to the expression levels of regulated cAMP, PKA, AQP5, CREB gene, increased airway fluid secretion related. Tip cAMP, PKA, AQP5, CREB gene locus may be the key target of lingganwuweijiangxin decoction treated asthma;4.Lingganwuweijiangxin decoction rises the expressions of cAMP, PKA AQP5 CREBproteins upregulate of rats lung, and it can reduce the airway inflammation, treat asthma. lingganwuweijiangxin decoction treat effects in the treatment of asthma is to increase the expressions levels cAMP, PKA, AQP5, CREB protein.
