Objective: Skin grafts have good treatment effects for skin damage caused by trauma, but a variety of factors has a great influence on the flap survival, the establishment of a network of blood vessels in the subcutaneous tissue has a decisive role in survival. Recent studies have proved that platelet rich plasma can release a variety of growth factors, and has a great role in promoting the formation of vascular endothelium. Current research studied the platelet rich plasma through the animal experiment on big white rabbit skin transplantation survival rate, to further clarify the role of platelet rich plasma gel on wound healing, in order to promote in the clinical use. Method:1. Animal experiment preparation: choosing 15 big white rabbits as experimental research subjects from animal research center of yanan university school of medicine. 1d before the preparation of animal models,we extracted 10 ml of arterial blood to prepare platelet-rich plasma from the 15 big white rabbits ear central artery respectively, and fully automatic blood analyzer was developed for the determination of platelets in platelet rich plasma concentration. The left and right sides of each big white rabbit’s back were treated with surgery to make models, and the left side was the experimental group, then treated with rich platelet gel skin, and the right side was blank control group, treated with saline leather piece. Were moved animal hair in the preparation of animal models before 1 day of skin surgery.2. Experimental animal model preparation: we cut for about 3cm diameter with full-thickness skin dermal vascular net work with surgical blades in 15 big white rabbits’ back left and right sides of the corresponding area. Experimental group wounds were treated with platelet-rich plasma and skin in situ implantation, and pouch fixed. Control group wounds were treated with regular cleaning, skin in situ implantation, and pouch fixed.3. Experimental observation: we observed skin survival situations in 7 days after regular demolition of flap pockets. And we randomly selected five big white rabbits in 7d,12 d,14d after the operation, and the central part of full-thickness skin tissues of experimental group and control group were treated with path ological measurement, and observed fibroblast, observe skin survival situation, inflammatory cells, etc. Result:1. White rabbit whole blood platelet count was(223.47 ± 13.03) × 109 / L, and platelet count in the platelet rich plasma through the secondary centrifugation was(983.73 ± 16.68) × 109 / L, and reached four to five times the concentration of whole blood platelet recommend by Marx.2. We observed flap survival situation in 7d after open the outside flap pockets, and found that the experimental group skin graft survival rate was(87.03±2.97) %, blank control group survival rate was(78.12 ±4.54) %, t = 6.36, P < 0.05. The platelet rich plasma treatment of skin flap survival rate was higher than commonly saline treatment, the difference of two groups’ data was obvious and has statistical significance.3. The experimental group biopsy has a lot of fibroblasts and lymphocytes in 7d after the operation, and the blank control group was less.We found the fiber cells and lymphocytes decreasedin12 d, and capillary number increased obviously. And the blank control group capillary number is less, the rest of the cell in 12 d is more than 7 d. The experimental group biopsy was found that fibroblast cells, lymphocytes, capillaries decreased in 14 d, granulation tissue hyperplasia, obviously with the increased number of collagen fibers. And the change of blank control group fiber cells, lymphocytes and blood capillary is not obvious, few number of collagen fibers in 12 d. Conclusion:1. Autologous platelet rich plasma can obviously increase the survival rate of the dermal vascular network skin graft in white rabbit.2. Autologous platelet rich plasma can obviously increase the proliferation of fibroblastcells,lymphocytes,capillaries and collagen fibersin the dermal vascularnet work skin graft of white rabbit.
