Objective Evaluate the significance of letrozole-induced polycystic ovary syndrome rat models. Explore the serum level of irisin,sex hormones and insulin resistance in PCOS obesity, PCOS non-obese group, the non-PCOS obesity group, the non-PCOS non-obese group; and analyze the movement on them. Analysis of the gastrocnemius FNDC5 m RNA expression in each group. Research correlation of serum levels of Irisin between groups before and after exercise rats model.Methods PartⅠ To feed 6-week female SD rats with four different conditions:A feed with ordinary feed, water and 1%CMC;B feed with ordinary feed, water, The concentration of 1.0mg/kg.d letrozole and 1%CMC;C feed with high fat feed, water and 1%CMC;D feed with high feed, water, The concentration of 1.0mg/kg.d letrozole and 1%CMC to built four different rat models.Part Ⅱ PCOS obesity(n=15, rejecting seven weight does not conform to the requirements), and PCOS non-obese(n=22), the non-PCOS obesity(n=16, rejecting six weight does not conform to the requirements), the normal weight PCOS(24) is further divided into exercise group and the non-exercise group(A-H). exercise groups run on wheel running machine for 3 weeks, non-exercise groups as control group. The HE staining was used to observe the ovary morphology; serum T, LH and FINS levels of each groups were measured by radioimmunoassay technique; FPG was measured by glucose oxidation technique; serum Irisin concentrations were measured by Elisa; Rt-PCR method was used to detect FNDC5 m RNA expression in rat gastrocnemius muscle; analyse and compare differences above of each groupsResult PartⅠ:1.(1)Estrus recovery time: PCOS obesity exercise group and PCOS obesity non-exercise group, the normal weight PCOS exercise group, the PCOS than normal body exercise group were 9.5, 13.6, 6.3, 10.5 days.(2)The LH and T levels of PCOS obesity exercise group and PCOS obesity non-exercise group, the normal weight PCOS exercise group, the PCOS than normal body exercise group were lower after exercise, p < 0.05, the difference was statistically significant.The INS and FPG of each group,were lower after exercise,but the exercise groups drop more, with the exception of the non-PCOS normal weight exercise group and the non-PCOS group normal body weight non-exercise group.(3)LH and T value were reduced after motion,in PCOS group the exercise group were reduced more than non-exercise group, p < 0.05, the difference was statistically significant.(4)HOMAR-IR: After movement each group were decreased, but compared with the non-exercise group, there is no statistically significant(p > 0.05).(5)IRISIN: PCOS normal weight exercise group is higher than PCOS normal weight non-exercise group, the non-PCOS obesity exercise group is higher than non-PCOS obesity non-exercise group, PCOS obesity exercise group is higher than PCOS obesity non-exercise group, p < 0.05, the difference was statistically significant.2.Correlation analysis showed that: IRISIN levels of obese after exercise were negatively correlated with INS, FPG. IRISIN of PCOS group after exercise were LH, INS, and weight were positively correlated, but negatively correlated with FPG and HOMAR- IR.3. In multivariate linear regression analysis: LH and INS are significant predictors of PCOS group before exercise β= 0.6560, 0.340(p < 0.005). after exercise, body weight and HOMAR- IR predictor is meaningfulβ= 2.436, 1.056, 1.228(P < 0.005);The non-PCOS group before exercise INS are significant predictors ofβ= 0.930(p < 0.001);After the PCOS group movement without significant predictor;4. FNDC5 m RNA expression:The RT-PCR values between exercise and non-exercise have statistical difference, p < 0.05. PCOS group rats FNDC5 m RNA value was lower than those of the non-PCOS, and the value of the obese was lower than non-obese group.5. Ovary viscera index and weight: PCOS group were smaller than the non-PCOS group, p < 0.05, the difference was statistically significant; obese group were smaller than normal weight group, p < 0.05, the difference was statistically significant.The exercise group is bigger than the non-exercise group, p < 0.05, the difference was statistically significant.Conclusion 1. Used letrozole and high-fat feed to feed SD rats for building PCOS and obesity models, were suitable for study the endocrine change.2. Exercise can reduce LH, T, INS, FPG of the PCOS obesity and PCOS non-obesity rats, but can increase its IRISIN values.IRISIN values in serum may Have an impact on treatment and outcome of PCOS, obesity.
