Objective:The aim of the study is to investigate the association between circulating tumor cell numeration and clinicopathological parameters in patients with colorectal cancer and further analyze the diagnostic value and advantage of circulating tumor cell compared with traditional biomarkers.Methods:Between July 2015 and October 2015,61 newly diagnosed patients with colorectal cancer were enrolled in this study, a total number of 20 patients with benign disease and healthy volunteers were used as control group.Each individual including control group had a 7.5ml peripheral blood samples taken. Immunomagnetic beads coated with tumor cell-specific Ep CAM antibodies were utilized to enrich circulating tumor cell from 7.5ml of blood samples,then the enriched samples will be further immunofluorescently stained.Clinicopathological factors including age, sex, site of primary tumor, TNM stage and the level of traditional biomarkers were collected. Assessment of the advantage of circulating tumor cell were also performed.Results:1、No association between pretreatment level of circulating tumor cell and clinicopathological parameters are observed(P>0.05).2、Pretreatment level of CEA are associated with N status,M statues and TNM stage(P=0.001,P=0.001,P=0.007).So is CA199(P=0.007, P=0.001,P=0.001).No association between the two biomarkers and T status were observed(P=0.06 and P=0.09,respectively).3、In 32 patients with negative CEA,18 of them were detected circulating tumor cell and12 patients are stage Ⅰor Ⅱ.In 43 patients with negative CA199,27 of them were detected circulating tumor cell and 15 patients are stage Ⅰor Ⅱ.18 patients with normal CEA and CA199 level were detected circulating tumor cell,and 12 of them are stage ⅠorⅡ.Conclusion:1、There is no association between pretreatment level of circulating tumor cell and clinicopathological parameters in patients with colorectal cancer.2、N,M and clinical stage are associated with the positivity of traditional biomarkers.3、Circulating tumor cell combined with traditional biomarkers can be served for early diagnosis in patients with colorectal cancer.
