Objective:To investigate the expression of yes-associated protein, p53 gene, the cell apotsis level,and the relationshipamong them in oral squamous carcinoma.Explore the mechanism of OSCC.Discovering a new target for OSCC therapies.Methods:1. Immunohistochemistry(IHC) was used to detect the expression of YAP and p53 in10 specimens of normal oral mucosa and 60 specimens of oral squamous carcinoma, Kapla n–Meier curves, log-rank tests, and Cox regression were used to summarise associateons between biomarker expression and overall survival.2. Westen Blot was used to detect the difference of protein YAP in normal oral mucos-a and oral squamous carcinoma.3. Apoptosis was observed by terminal deoxynucleotidyltransferase-mediated d UTP nick-end labeling( TUNEL) in the 10 specimens of normal oral mucosa and 60 specimens of oral squamous carcinoma.Results:1. YAP was not expressed in normal oral mucosa epithelium or only expressed in basal cell nucleus. The difference of YAP expression level between cancer and normal tissue was significant(P < 0.05). Western blot results show that the expression level of YAP in cancer and normal tissue was ignificant(P < 0.05). Expression of YAP migrated from nucleus to cytoplasm when the differentiation degree decreased. Besides YAP was also associated with differentiation degree, lymph node metastasis and TNM staging(P <0.05), and positive rate was increased as the malignant degree enhanced.2. p53 protein was not expressed in normal oral mucosa or only a few expressesd in basal nucleus, p53 was expressed in OSCC cell nucleus. The difference of p53 expression in normal tissues and the cancerous tissue were significant(P < 0.05). The difference of p53 expression in high, medium and low differentiation were significant(P < 0.05), and the expression of p53 gene is associated with lymph node metastasis and TNM staging(P< 0.05).3. The difference of apoptosis level in cancer and tissues were significantly(P < 0.05),and also in the high, medium and low differentiation(P < 0.05). Apoptosis level decreases with degree of differentiation.in addition, the level of apoptosis associated with lymph node metastasis and TNM staging(P < 0.05).4. The correlation between YAP, p53 and YAP, cell apoptosis were positive(r = 0.343,P < 0.05) and negative(r =- 0.402, P < 0.05), there was no relationship between apoptosis level and p53 gene.5. The survival rates of YAP positive and negative group were 76.0%, 90.0%, the difference was statistically significant(P < 0.05); In multivariable analyses, YAP was the independent factor of prognosis of OSCC(P < 0.05).Conclusion:1. Expression of YAP in OSCC increased obviously, and related to the expression of p53.Both of them participated in the occurrence and development of OSCC.2. The correlation between YAP and cell apoptosis was negative, inhibiting the functi-on of YAP to promoting apoptosis has a certain value in the treatment of OSCC.3. High expression of YAP was closely related to the prognosis of OSCC patients. It has a certain instructin in assessing patients’ survival risks, improving the patients’ survival rate and the quality of life.
