Research On The Change Of IL-10 To Severe Acute Pancreatitis Complicated With Renal Injury In Rats In Different Altitudes

Objective: By establishing severe acute pancreatitis(SAP) rat models in different altitudes, analyzing the relationship between SAP complicated with renal injury and IL-10 from perspective of cytokine.Methods: A total of 160 male SPF Wistar rats were selected as research object and divided into Xi’an(A)(altitude,1027m), Xining(N)(2260m), Xinghai(X)(3300m), and Wenquan(W)(3950m) group, and each group was further divided into 5 groups(1h, 6h, 12 h, 24 h and F group), with 8 rats in each. SAP rat models were established by injecting sodium taurocholate via capsula pancreatic, and rat models in control group were established by flipping the pancreas. Blood from abdominal aorta and renal tissue were obtained at corresponding time point. Blood amylase, creatinine, and blood urea nitrogen were tested by centrifugation method, and expression of IL-10 in renal tissue was evaluated. The data were presented as mean ± standard deviation, one-way ANOVA was employed to compare the data within group and among groups, S-N-K was used to make pairwise comparison, and pearson correlation was used for pathological scoring as well as correlation analysis. Inspection level was set as α = 0.05.Results: 1 Change of serum amylase. For SAP groups in the same altitude, P < 0.05,and difference was statistically significant. Compared with SAP groups, change of serum amylase in F groups was more obvious, P < 0.05,and difference was statistically significant. For SAP groups in different altitudes, serum amylase changed as altitude elevated, P < 0.05, and difference is statistically significant. Difference between serum amylase of F groups in different altitudes was not statistically significant,P > 0.05. 2 Change of serum creatinine and blood urea nitrogen. For SAP groups in the same altitude, P < 0.05, and difference was statistically significant. Compared with SAP groups, change of serum creatinine and blood urea nitrogen in F groups were more obvious, P < 0.05, and difference was statistically significant. For SAP groups in different altitudes, serum creatinine and blood urea nitrogen increased as altitude elevated, P < 0.05, and difference was statistically significant. Difference between serum creatinine and blood urea nitrogen of F groups in different altitudes were not statistically significant,P > 0.05. 3Pancreatic tissue pathological score. For SAP groups in the same altitude,the elevation of the pancreatic tissue in SAP group score in comparison to the different time, P < 0.05, and difference was statistically significant. SAP group and F group comparison, the score difference, P < 0.05, statistical significance; with the increase of altitude, at the corresponding time pathology score gradually increased, P < 0.05, the difference is statistically significant; compared different altitude Group F pancreatic histologic score, P > 0.05, the difference was not statistically significant. 4Change of IL-10 expression in renal tissue. For SAP groups in the same altitude, P < 0.05, and difference was statistically significant. Compared with SAP groups, change of IL-10 expression in F groups was more obvious, P < 0.05, and difference was statistically significant. For SAP groups in different altitudes, IL-10 expression increased as altitude elevated, P < 0.05, and difference was statistically significant. Difference between expressions of IL-10 of F groups in different altitudes was not statistically significant, P > 0.05. 5 At the same altitude, there was significantly correlated with the level of P<0.01 in the expression of IL-10, AMY, Cr, BUN, and pancreas pathological grading. Statistical data at the same time points were significantly correlated at the level of P<0.01 or P<0.05; there was no correlation between the statistical data of the F group.Conclusion: 1High plateau environment with low oxygen can aggravate SAP complicated with renal injury;2 IL-10 increased with the development of severe acute pancreatitis complicated with renal injury;3 There were significantly correlated for SAP groups with the pathology scor of pancreatic tissuee, blood AMY, Cr, BUN and the expression of IL-10 in renal tissue.
