Objective: This article eatablished rat’s SAP model by animal experiment,analyzing the changes of various biochemical indexes and theexpression of seriously related factor: thromboxaneA2(TXA2) in renalinjury, we investigate the degree of renal injury during the differentprocess and development of SAP.That is to provide a new theory ofbasement for the clinical diagnosis and therapy of SAP.Material&Methods:72specific pathogen free (SPF) male wistar ratswere divided into three groups randomly: Blank group (B group, n=24),Sham-operation group (F group, n=24), SAP group (SAP group, n=24),The each group were randomly divided into three groups:6h,12h, and24h. Each group included8rats. B group do nothing before were killedat6h. F group only flipped and strike pancreas about6-10times.SAPgroup was touched by the pancreas under the injection of5%sodiumtaurocholate (Na-Tc)(1ml/kg), in order to establish an animal model ofSAP, after the models successfully established,under6h,12h,24h rats werekilled; All rats were collected serum, pancreas and kidney tissue. Thecontent of serum amylase (AMY) and the biochemical indexes of renalfunction were observed by automatic biochemical analyzer. Observed thepathological changes of pancreatic and kidney tissue after HE staining.Test thromboxaneA2in serum and kidney tissue with solid phasesandwich enzyme-linked immunosorbent assay (ELLSA) Results:SAP model was successfully established; The results ofpancreasous and renal pathology score, The results of content of serumamylase statistics, The results of biochemical indexes of renal functionstatistics, The results of the concentration of thromboxaneA2(TXA2) inserum and renal tissue analyze:①Comparison between groups: comparedwith blank group, in the different time points,false surgery group has nostatistical significance(P>0.05),and the SAP group was of statisticalsignificance(P <0.05);compared with false surgery group, in thedifferent time points, SAP group all were of statistical significance(P<0.05).②Comparison in the group: in the different time points, the blankgroup has no statistical significance(P>0.05),and the false surgerygroup has no statistical significance(P>0.05) and the SAP group allwere of statistical significance(P<0.05). The concentration ofthromboxaneA2(TXA2) in serum is related positively with renalpathological score(r=0.928, P<0.01), the concentration ofthromboxaneA2(TXA2) in renal tissue is related positively with renalpathological score(r=0.883,P <0.01),renal pathological score is relatedpositively with pancreasous pathology score(r=0.981,P <0.01),theconcentration of thromboxaneA2(TXA2) in serum is related positivelywith pancreasous pathology score(r=0.934,P <0.01),the concentrationof thromboxaneA2(TXA2) in renal tissue is related positively withpancreasous pathology score(r=0.891,P <0.01).Conclusions:1.Kidney injury relates to the degree of severe acutepancreatitis in severe acute pancreatitis.2. The obvious change of TXA2in kidney tissue closely associated with the degree of kidney injury insevere acute pancreatitis.
