| Objective:To investigate the effects of limited fluid resuscitation on the Lung tissue morphologyinã€inflammatory response and apoptosis in rats with uncontrolled hemorrhage at the early stage.Mainly according to finding out the differences of the level of pathological examination of HE-stain ã€the expression of NF-кB and ICAM-1ã€the situation of cell apoptosis between different ways of fluid resuscitation. We can get a conclusion that the limited resuscitation can contribute to the protection of the lung, from which we can get a pathophysiological theory which can help building the theoretical foundation of the clinical practice. Methods: Sixty SD rats were equally randomized into 4 groups: blank control group(C),limited resuscitation group( LR), conventional fluid resuscitation group(TR) and non-resuscitation group( NR),each group contains 15 rats, after being resuscited, they would be killed after 2 hours. The ultrastructure in lung tissue was observed by electron microscope, the protein levels of NF-кB in lung tissue was determined by enzyme-linked immunosorbent test, immunochemistry staining was used to detect the expression of ICAM-1 on lung while the apoptosis of lung was tested by means of TUNEL.Results:When the resuscitation was completed, the level of pathological examination of HE-stain ã€the expression of NF-кB and ICAM-1ã€the situation of cell apoptosis show that the limited resuscitation group(LR)and traditional resuscitation group(TR)were lower than none resuscitation group( NR) while apparently higher than the control group(C)(P<0.01);LR group was lower than TR group on the pathological examination of HE-stain and the reflexing of cell apoptosis(P<0.01),and is also lower on the expression of NF-кB and ICAM-1(P<0.05).Conclusion:The limited fluid resuscitation can help reducing the responding of NF-кB and ICAM-1,releasing the pathological damage of lung tissue and the level of cell apoptosis, which may play the role of protecting the lung from further damage. |
