Objective PVC(Polyvinyl chloride, PVC) is a kind of widely used synthetic material, data show that the national production of Polyvinyl chloride resin is 13.1776 million in 2012.Vinyl Chloride Monomer(VCM) is a kind of important chemical raw materials, mainly used in the synthesis of polyvinyl chloride(PVC) and the plastics industry. A large number of studies have shown that long-term exposure to vinyl chloride can produce toxicity to multiple organ systems.VCM is one of the certain human carcinogenic agents, can lead to multiple organ tumors in many systerms. The carcinogenic mechanism study is mainly on the hypothesis that VCM active metabolites cause genetic material damage and start the carcinogenic process. VCM occupational exposure limits in developed countries is 1 PPM(2.79 mg/m3, the PEL) at present, the VCM time weighted average of the allowable concentration(PC- TWA) in our country is 10 mg/m3, short time contact allowable concentration(PC- STEL) is 20 mg/m3, are several times higher than western country health standard.Previous studies mainly analyze the relationship between gene polymorphism and VCM injury, only looking for susceptibility biomarkers from the perspective of gene polymorphism. There are few practitioners to analyze it from the level of protein—the products of gene, the real executant of life functions. Many protein biomarkers show the realization of function or the development of disease by change of content of abundance.Therefore, it’s very important to use proteomics to measure the relative and absolute concentration of protein. Our study using i TRAQ combined LC-MS/MS technology screen the serum concentration of differential protein measured in workers with different VCM exposure and analyze the function of differential protein. It provides a certain basis for clarifying the pathogenesis of disease resulted from VCM exposure and finding the biomarker of VCM exposure.Methods According to the data of factory’s layout chart and manufacturing technique provided by its own security section, we get the plant’s basic production situation data and determine the main contact workshop.We start field investigation on production building and production equipment, found production chain and location that may lead VCM escape. Talk to the workers and do questionnaire survey at the same time in order to understand working type, working system and working manner of each workshop.Objects of this study included 135 cases, all of them come from the same chemical company in Tianjin. It contacts 112 VCM exposed workers and 23 blank controls. Fasting venous blood was collected from 23 healthy controls and 112 VCM-exposed workers(29 people, low-dose exposure, 22 people, mid-dose exposure,61 people,high-dose exposure) according to accumulated exposure doses. Removal of albumin and globulin, proteins were precipitated by acetone precipitation. After digestion and i TRAQ labeling, these proteins were identified by 2D-nano LC-ESI-TOF/MS.Result We use occupational hygiene investigation method to evaluate and analyze the status quo of occupational exposure. By Proteomics research method-i TRAQ,we totally defined 248 kinds of proteins, including 216 quantify proteins. This experiment found out 11 differential proteins, such as C4b-binding protein beta chain、Coagulation factor V 、 Hemoglobin 、 Haptoglobin-related protein 、 Keratin 、 Cadherin 5 、Transforming growth factor 、Homo sapiens pregnancy-zone protein and so on, which including 7 up-regulation proteins and 4 down-regulation proteins.Conclusion We should carry out prevention and control method of occupational hazard according to the characteristics of PVC production enterprises, improve the occupational hazard protective measures in order to preferably protect laborers’ health. Our experiments aimed at studying serum of those who are exposed to different doses of vinyl chloride. It turned out that the expression of tumor associated proteins do not appear obvious trend among vinyl chloride doses. This may due to the occurrence of tumor requires higher cumulative exposure dose. But to a certain extent, the experiment clues to mechanisms that how VCM induce tumors and has certain reference value in finding the protein biomarkers contacted with VCM.
