Objective In this study,we preliminary analyzed the differentially expressed proteins in synovial tissue of patients with RA?OA and normal trauma by i TRAQ technique,and investigate the correlation of protein expression in metabolic pathways and these differences in the pathogenesis of RA,and provide a theoretical basis for the formation mechanism and clinical intervention on RA.Methods To collect the RA?OA and normal trauma patients,synovial tissue after surgery abandoned from March 2015 to July 2016 in Lanzhou University as a research object,The protein was analyzed qualitatively and quantitatively by using i TRAQ technique.GO function analysis and KEGG pathway analysis were performed on differentially expressed proteins.Results 1.Quantitative analysis of differentially expressed proteins showed that395 differentially expressed proteins were found in RA group compared with N group,there were 216 up-regulated proteins and 179 down-regulated protein;RA group compared with the OA group,There were 421 differentially expressed proteins,up-regulated protein were234,down-regulated protein were 187;The OA group compared with N group were found total of 326 differences expression protein,132 differentially expressed proteins were up-regulated,94 differentially expressed proteins were down–regulated.2.The GO protein function annotation of differences expressed proteins: RA group compared with the N group found that there are 24 kinds of biological process function classification,there are 11 kinds of molecular function,and located in 10 kinds of cells;RA group compared with OA group showed that there are 25 different biological processes?11 molecular functions and located at 10 cell types.And the OA group compared with N group,there are 24 kinds of biological processes functional classification,12 kinds of molecular function,located in 11 kinds of cells.3.Through to the differentially expressed proteins KEGG Pathway analysis:RA group and N group?RA and OA?OA group and N group,there were 35?41and 20 signaling pathways respectively.Conclusion 1.According to protein family,FIB family involved in the occurrence and development of RA,is expected to become new molecular markers of RA diagnosis.2.The level of MMPs and CRP protein were correlated with the disease activityof RA patients.3.PREX1?APO C-I may be involved in the development and progression of RA disease.4.PI3K-AKT signaling pathway has potential value in the treatment of patients with RA.
