A Study On Antitumor Function Of Huajiyiganjian In H₂₂ Tumor Bearing Mice By Expression TGF-β1 P57^kip2

Objective : Mouse models of solid tumor H22 liver cancer were established.The expressions of TGF-βl and P57kip2were detected by immunohistochemistry. The mechanism of Traditional Chinese medicine can inhibit TGF-βl expression and improve expression of P57kip2 in The H22 cells of liver carcinoma.Methods:(1)The H22 cells of liver carcinoma were (inoculated) in KM mouse armpit hypodermically.(2)A total of 60 mice with H22 cells of liver carcinoma were randomly divided into physiological saline group , TCM group,5-FUO group and TCM +5-FUO group(.3)The expressions of TGF-βl and P57kip2 were detected by immunohistochemistry.Mesults:(1)evaluation synthetically the effect of TGF-βl by index systemTGF-βlwas mainly expressed in the cytoplasm and partially in the nucleus of tumor cells.The expressions of TGF-βl are higher in the physiological saline group than in any other group(p<0.05).There is no obviously differences between the TCM groupand 5-FUO group(p>0.05)The expressions in TCM +5-FUO group is much more lower than any group of the experiment.(p<0.05)(2)evaluation synthetically the effect of P57kip2 by index system Observed under optical-microscope,the signal of P57kip2 is sepia or brown,mainly in the nucleus of tumor cells , partially in the cytoplasm.The expressions of P57kip2 are higher in the TCM +5-FUO group than in any other group(p<0.05). There is no obviously differences between the TCM groupand 5-FUO group(p>0.05).The expressions in physiological saline group is much more lower than any group of the experiment(p<0.05). Conclusiont : Tumor is a kind of disease that related with gene. activation of proto-oncogenes.The devitalization of tumor suppressor genes and the disorder in signal transduction system result in cell proliferation and apoptosis.Traditonal Chinese Medicine can inhibit the tumor cell effectively..The Mechanism of this procedure is base on improve the expression in tumor suppressor genes and inhibit proto-oncogenes.Compare with Western Medicine cancer Treatment,the clinical effect was satisfied. Chinese-western-combined therapy can be choosed for the cases according to the patient's condition.
