| [Objective] To investigate the role of chemokine CCL19 and chemokine receptor CCR7 in invasion and metastasis of different types lung cancer cell.[Methods] (1) The expression of chemokine receptor 7 were detected by immunofluorescence experiment in lung cancer cell line A549, EPLC, GLC, YTMLC and XLA-07.(2) The cell proliferation were explored by CCK8 in different lung cancer cell lines.(3) The invasion ability were tested by transwell chamber system in different lung cancer cell lines and the influence of CCL19, CCR7 neutralizing antibody in invasion ability of different cell types. The groups were divided into:A549, A549 (L), A549 (L-RmAb), EPLC, EPLC (L), EPLC (L-RmAb), GLC, GLC (L), GLC (L-RmAb), YTMLC, YTMLC (L), YTMLC (L-RmAb), XLA-07, XLA-07 (L), XLA-07 (L-RmAb) groups.(4) The migration ability were tested by cell scratch experiment in different cell types. (5) The expression of CCR 7 was detected by real-time fluorescence quantitative polymerase chain reaction in different lung cancer cell lines and clinical samples. (6) The expression of CCR7 was analyzed by immunohisteochemical staining in clinical samples. (8) SPSS 17.0 statistical software was used for data analysis.[Results] (1) There were CCR7 expression in lung cancer cell line A549, EPLC, GLC, YTMLC, XLA-07 and the positive signal were located in the cytoplasm and cell membrane.(2) Chemokine CCL19 had obvious effect on promoting lung cancer cells proliferation, however, CCR7 neutralizing antibody can inhibit the ability of CCL19 promoting proliferation (3)Number of invation cells is significantly increased after CCL19 treatment, while the number of invation cells had no appreciable increase after treatment with CCL19 and CCR7 neutralizing antibody. (4) The migration ability of lung cancer cells had significantly increased after CCL19 treatment; while the migration ability of lung cancer cells had no appreciable increase after treatment with CCL19 and CCR7 neutralizing antibody. (5) Real-time fluorescent quantitative PCR:CCR7 was expressed in 5 kinds of lung cancer cells and the CCR7 expression amount was A549>EPLC>YTMLC>GLC> XLA-07 from strong to weak. CCR7 expression in lung cancer tissues was higher than that in normal lung tissues. (6)Immunohistochemical:there was the CCR7 expression in different regions and pathological types of lung cancer tissues. The CCR7 expression increased significantly in xuanwei and gejiu of lung cancer tissue than other areas of lung cancer tissue, while there was no obvious difference between Xuanwei an Gejiu region. The expression of CCR7 was related to lymph node metastasis, which is not related to the pathological type of lung cancer.[Conclusions] (1) There was CCR7 expression in a variety of lung cancer cell line and lung cancer. (2) Chemokines CCL19 and receptor CCR7 are key factors in the regulation of invasion and metastasis of lung cancer cells. (3) CCR7 expression had regional differences, and its expression was related to lymph node metastasis, which is not related to the pathological type of lung cancer. |
