Objective:Inhaled corticosteroids(ICS) are the most effectively and commonly used controller medications prescribed for asthma. Nevertheless, asthmatic response to corticosteroids also varies widely between individuals, some people even have steroid-resistance. The individual differences is associated with asthma -related genes. Therefore, this study will investigate the association between GLCCI1 rs37973 polymorphism and the effect of inhaled budesonide in asthma, and also to observe the asthma symptoms, lung function and the count of peripheral eosinophil in different phenotypes of GLCCI1 polymorphism in asthma,to understand the correlation between GLCCI1 polymorphism and clinical phenotypes of asthma.Methods:Forty-five asthmatic with mild persistent asthma of The First Affiliated Hospital of Kunming Medical University from January 2015 to December 2015 were included, genomic DNA from peripheral blood were extracted for single nucleotide polymorphism analysis by fluorescence staining in situ hybridization,at the same time,the percentage of peripheral blood EOS(ESO%) were conducted.All subjects inhaled budesonide suspension liquid lmg once daily for 8 weeks. Before and after 8-week treatment,asthma symptom scores,lung function and histamine bronchial provocation test were measured.All statistic was performed using SPSS20.0. A basic description of measurement data was expressed as mean±SEMs, the enumeration data adopts with ratio and composition, the assumption checking of measurenment data was expressed as paired t-test.The count data was applied to c2-test. Significance was accepted at a P value of less than 0.05.Results:1. All subjects presented polymorphism in GLCCI1 rs37973,showed as AA genotype,AG genotype,GG genotype.The frequencies of AA、AG、GG genotype in asthmatic were 31.1%、51.1%、17.8%,respectively.2. The association between GLCCI1 polymorphism and clinical phenotype of asthma: The gender,age and BMI were not different significantly in all genotypes (P>0.05).The PD20-FEV1 in asthmatic AA genotype was (2.08±0.28) ummol/l,significantly lower than that in asthmatic AG、GG genotype [(2.93±0.35、2.82±0.29, respectively)ummol/l](P=0.002), indicated the airway hyperresponsiveness in asthmatic AA was higher AG、GGThe EOS% in asthmatic AA was(5.30±1.01)%, significantly higher than that in asthmatic AG、GG(3.49±0.52、4.12±1.08, respectively)%] (P<0.001). There is no differetion in daytime symptom scores,night symptom scores,FEV1,FEV1/Pred%,FEV1/FVC in all genptypes(P all>0.05).3. The association between GLCCI1 polymorphism and the of inhaled budesonide.① The association between GLCCI1 polymorphism and the improvement of ashma symptom scores in response to inhaled budesonide: The daytime symptom scores and night symptom scores of all subjects improved significantly after 8-week inhaled budesonide treatment (P all<0.05).But there was no significantly difference in the improvement of both daytime and night symptom scores in three genes asthmatics(P>0.05).② The association between GLCCI1 polymorphism and the improvement of pulmonary function in response to inhaled budesonide: The FEV1,FEV1/Pred% improved signicantly after treatment in all subjects,but there was difference in different genotypes.The improvement of FEV1,FEV1/Pred% in AA was significantly higher than AG/GG(P<0.05).③ The association between GLCCI1 polymorphism and the improvement of airway hyperresponsiveness in response to inhaled budesonide: The PD20-FEV1 improved significantly after treatment in AA genotype asthma,indicated the airway hyperresponsiveness improved significantly(P<0.05),and average increasing levels significantly higher than in AG、GG.Conclusions:The GLCCI1 rs37973 locus polymorphism is maybe associated with the pesponse of inhaled budesonide in asthma.
