Objective:To investigate the effect of JianPi ShuGan Kang Du decoction on CXCR4/CXCL12 axis of the triple-negative breast cancer (TNBC) ratsMethods:Human breast cancer cells (MDA-MB-231) were transfected with pAcGFP-C1 by Fugene HD Transfection Reagent. The cells which express the pAcGFP gene were selected by geneticin (G418).The percentage of MDA-MB-231-pAcGFP was detected by Flow cytometry(FCM).The mice bearing MDA-MB-231-pAcGFP cells were obtained with subcutaneous inoculation.The volume and the weight of the xenograft were observed.At the end of the experiment,the metastasis in the lung,liver and kidney were detected by the fluorescence imaging.Examine cells in tumour microenvironmen by Immunohisochemical staining and using Western Blot to test the expression of association protein(including CXCR4,CXCL12,HIF a, NF-κB,VEGF).Results:The rate of the MDA-MB-231-pAcGFP cells was 96.5% and the rate of tumor formation with these cells was 100%.The average weight of the xenograft was 1.30±0.11g (group A),0.75g±0.12g(group B),0.48g±0.09g (group C) and 0.44g±0.07g(group D) respectively,The volume of these xenograft changed accordingly, they were 6.635± 0.5cm3,4.017±0.466 cm3,2.664±0.7 cm3 and 2.323±0.492 cm3 respectively (compared with the control group,p< 0.05). The tumor inhibition rate of the three groups were 42.18%,62.95%, 66.28%,respectively. Metastasis in the liver were found in the group A and group C and no metastasis was found in the group B and group D. JianPi ShuGan KangDu ricipe can significantly suppress the infiltration of tumor associated macrophages(TAMs) and cancer-associated fibroblasts(CAFs), and ADM3100 can enhance that effection. Protein expression of group B, C and D are inhibited with varying degrees, also we found group D has a higher inhibition ratio.Conclusion:JianPi ShuGan KangDu decoction can regulate CXCR4/CXCL12 axis related molecules expression through changing the tumor microenvironment, so that it could inhibit the growth and liver metastasis of human breast cancer xenograft in nude mice.
