Research On Effect Of Guiluerxianjiao On The Osteoporosis In Adult Zebrafish

Objective:In our study Zebrafish was selected as model animal to provide new idea of osteoporotic model.To observe the effects of Gui lu Er xian Glue on the changes of bone metabolism and microstructure on zebrafish osteoporosis modle,and observe the structure of Type I collagen,try to reveal the mechanism and provide experimental basis for clinical use of Gui lu Er xian Glue for the treatment of OP.Methods:(1)Adult Zebrafish were exposed to a serial concentrations of prednisolone,and Ultrapure water was selected as the vehicle control group. All groups were incubated in cistern (28.5℃) for two weeks. Zebrafish bones after two weeks were stained with HE,the effects of guiluerxianjiao in the staining area of scales and zebrafish head bones were investigated. Quantitative analysis of the stained area was performed by microscopic inspection and digital imaging methods to reflect the amount of bone.the content of Ca,P and ALP in the Zebrafish bones were measured by automatic biochemistry analyzer. (2)Adult Zebrafish were exposed to a serial concentrations of Guiluerxianjiao solutions with prednisolone,and Ultrapure water was selected as the vehicle control group. calcitriol was selected as the Positive control group.All groups were incubated in cistern (28.5℃) for two weeks. Zebrafish bones after two weeks were stained with Masson,the effects of guiluerxianjiao in the staining area of zebrafish bones were investigated, the structure of zebrafish bone collagen was observed by light microscope. Quantitative analysis of the stained area was performed by microscopic inspection and digital imaging methods to reflect the amount of bone.the content of Ca,P,ALP and Hyp in the Zebrafish bones were measured by automatic biochemistry analyzer.Results:(1)Compared with the blank group,with the increase of concentration of prednisolone,the cell number reduce, the content of Ca and P go down, the content of ALP go up. (2)compared with model group, under the microscopic, the collagen proteins increase significantly in the group with calcitriol and the serious groups with guiluerxianjiao.the content of Ca and P increase, the ALP reduce and the Hyp increase significantly in the group with calcitriol and the serious groups with guiluerxianjiao.Conclusion:zebrafish osteopenia model can be induced by prednisolone successfully.the Traditional prescription of guiluerxianjiao can tonifying Liver and kidney,then preventing against osteopenia, it Mainly promot osteoblast reproduction, promote the production of type I collagen, and mineralized bone, increase the bone density, improve osteoporosis.
