Objective:the tentative action mechanism of Xiezhikang capsules of Chinese medicine improving hypertension combined metabolic disorder is discussed by comparing body weight, blood lipid, blood glucose, serum insulin, visceral fat and proteinTyrosine Phosphatase-1B (PTP-1B) expression of rats under three feeding methods:normal feeding, high fat feeding and high fat+Xiezhikang capsules feeding.Method:Seventeen healthy rats were randomly divided into three groups:control group (STD, n = 5) group, high-fat group (HA, n= 6) and Xiezhikang group (HXZ, n=6), feeding basic feed, high fat feed+0.5% cellulose sodium and high fat feed+300 mg/(kg, d)Xiezhikang, respectively. Isodose 0.5% cellulose sodium and 300 mg/(kg, d)Xiezhikang perfused rat stomachs at HA and HXZ groups, respectively. Feeding for 8 weeks in the same external conditions and weekly monitoring body weight (BW). After 9 weeks, blood was extracted in mice eyes and used to test fasting blood glucose (FBG), fasting insulin (FINS), total cholesterol (TC), triglyceride (TG), serum insulin. Subsequently visceral adipose tissue weight of rats are measured, and body fat ratio calculation, HOMA index and Insulin resistance (IR) are calcuated. PTP-1B protein expression is analyzed by Western Blot method.Results:1. Weight of HA group rats increased significantly than that of STD group (P< 0.01). Comparison with STD group, Weight of HXZ group rats also significantly increased (P< 0.05), but is significantly lower than that of HA group rats (P< 0.05).2.High-fat feeding group rats fasting blood glucose, fasting insulin were significantly higher than that of normal control group rats (P< 0.01), HOMA index-IR was also significantly higher than that of normal control group (P< 0.05), capsule intervention group:rats fasting blood glucose, fasting insulin and HOMA IR index was significantly lower than high-fat feeding group (P< 0.05);fasting glucose intervention group and normal control group fasting glucose fasting insulin HOMA index-IR still significantly increased (P< 0.05), fasting blood sugar level decreased significantly (P< 0.01).3. High-fat feeding group rats epididymal fat weight, body fat ratio is significantly higher than the normal control group (P< 0.05), capsule intervention group and normal control group, high-fat feeding epididymal fat weight and body fat ratio had no obvious difference.4. PTP-1B expression level of rats at HA group is significantly higher than that of rats at STD group (P< 0.01). PTP-1B expression level of rats at HXZ group is lower than that of rats at HA group (P< 0.01), but is no difference with rats at STD group (P> 0.05).Conclusion:1.Chinese medicine Xuezhikang capsule can significantly lower the blood pressure after intervention combined metabolic disorder rat body weight, but the visceral fat and body fat than the improvement is not obvious.2.Xuezhikang capsule can obviously improve high-fat feeding sugar metabolic abnormalities and insulin resistance in rats.3.Xuezhikang capsule obviously improves insulin resistance for rats with high-fat feeding by down-regulating expression of visceral fat inhibits PTP-1B.
