Objective:The aim of this study was to stablish the T2 DM model, AS model and T2 DM + AS model. Then to discuss the relationship between Angptl4, Inflammatory factor with the atherosclerosis in type 2 diabetes mellitus with these models.Methods:20 ApoE-/- mice were randomly divided into control group, type 2 diabetes group, atherosclerosis group and type 2 diabetes with atherosclerosis group. The mice in control group were fed with normal diet and atherosclerosis group were fed with high fat diet. The mice in type 2 diabetes group was fed with nicotinamide +streptozotocin(STZ)+normal diet, while type 2 diabetes with atherosclerosis group was fed with nicotinamide +STZ + High- fat diet. The artery and pancreas were examined through normal method. The level of Angptl4, glucose, insulin, HOMA-IR, TG, PAI-1, LDL-c, inflammatory factorTNFα, TGF-β1 and CRP in serum were detected by ELISA, CLIA or biochemical detection. Immunofluorescence method was used to analysis the expression of α-SMA in artery in each group. The expression changes of Angptl4, α-SMA, VEGF, TNFα, TGF-β1 and CRP were detected by RT-PCR and Western Blot methods.Results: 1) The results of biochemical detection show that the content of Angptl4 in model group(T2DM group,AS group,T2DM+AS group)was lower than the control group, and T2 DM + AS group has the lowest content. Compared with the control group. The level of TNFαand CRP were increased while TGF-β1 was reduced significantly in the AS group and T2 DM + AS group, and more obvious change was seen in T2 DM + AS group than AS group. Insulin levels were reduced significantly in the model group, and more obvious phenomenon was discovered in T2 DM + AS group. HOMA-IR was higher in T2 DM group especially in T2 DM + AS group, TG, LDL-c and PAI-1 were higher in model groups, which was the highest in T2 DM + AS group.2)Compared with the control group,arterial intima narrow and overlap phenomenon were seen in T2 DM group and AS group, meanwhile, the thickness of artery intimal and the expression of α-SMA was increased in AS group. The atheromatous mottle was replaced by atheromatous plaque, and other phenomena were higher thanAS group significantly.3)Pancreas HE staining showed that compared with the control group, pancreas in AS group are relatively normal, but with inflammatory cell infiltration; T2 DM group summary interval inconspicuous pancreas, acinar atrophy vacuole formation; In T2 DM + AS group pancreatic lobules spacing is not obvious, acinar atrophy vacuole formation and inflammatory cell infiltration.4)The results of RT-PCR and Western blot show that, compared with the Control group, Angptl4 expressed in AS group and T2 DM group was significantly lower, and the AS group is lower than T2 DM group, T2 DM + AS group was the lowest. Compared with the Control group, the expression of TNF-α, TGF-β1 and CRP in T2 DM group did not change significantly; the expression of TNF-αand CRP in AS group increased while the expression of TGF-β1 is significantly reduced, T2 DM + AS group expression of TNF-α and CRP were significantly higher than AS group, the expression of TGF-β1 was significantly lower than AS group.Conclusions: 1) Theexpression of Angptl4 is lower in T2DM- AS group and it is related glucose metabolism and lipid metabolism indicators.2)Angptl4 could delay the development of atherosclerosis in type 2 diabetes mellitus through the regulation of inflammatory factor,so it is a protective factor for atherosclerosis in type 2 diabetes mellitus.
