Objective To evaluate and contrast the efficacy and safety for GLP-1 and metformin in patients of overweight or obesityMethods Systematically literature searching of CBM, VIP database, WFSD, EMBASE, PubMed, Medline, Cochrane Library databases and so on (the deadline is March,one,2016). We Selected the Randomized controlled experiments that the objects are overweight or obesity and using GLP-1 or metformin. And use the software of Stata 11.0 to analyse the data. Results At the end, we screen out 13 literatures, of them are randomized controlled experiments. Of all the objects, there were 1120 T2DM patients,134 T2DM nonalcoholic fatty liver patients,192 metabolism syndrome patients. The Meta-analysis showed:In the aspect of efficacy, The effect of GLP-1 on the improvement of the body weight, BMI and waist circumference is superior to metformin. The Combined effects are[SMD=-0.481,95%CI(-0.908,-0.053)]、[SMD=-0.925,95%CI(-1.272,-0.578)]、[SMD=-0.889, 95%CI(-1.720,-0.058)] respectively. In the aspect of safety, GLP-1 have higher incidence of adverse reactions. The Combined effect is [RR= 1.202,95%CI (1.082,1.336)].Conclusions The effect of GLP-1 on the improvement of the body weight, BMI and waist circumference is superior to metformin. However,in the aspect of safety, GLP-1 have higher incidence of adverse reactions.
