Objective How the caspase-1 inhibitor AC-YVAD-CMK neuro-protect EAE in Wistar rats and its mechanism.Methods 30 female Wistar rats were randomly divided into four groups:normal control group (NS,n=10), EAE control group (EAE+DMSO,n=10), AC-YVAD-CMK group (EAE+AC-YVAD-CMK, n=10). Wistar rats immunized with fresh guinea pig spinalcord homogeny eous. Then the AC-YVAD-CMK group was intraperitoneal injected with O.lmg/100g twice a day while normal control group and EAE group was saline until the rats get the Hooper score of 6 or 7,or were totally normal on immuned 21d. Observe the rats every day,pathology study the brain and spinal cord tissue after recod the score, and investigated the expression of the inflammatory factor of caspase-l,IL-1β,IL-18 by immunohistochemical staining.Results Normal group didn’t be infected while the incidence rate of EAE group and AC-YVAD-CMK group was 100%.The weight of nineteen rats decreased over time and one rate recovered.Compared with EAE group AC-YVAD-CMK group got delayed-onset,long latency and short time to Progress (p<0.05). Pathological changes:inflammatory cell didn’t fine in normal group,while that in EAE group is more than in AC-YVAD-CMK group.Caspase-l,IL-1β,IL-18 expressed more in EAE group than in AC-YVAD-CMK(p<0.05).Conclusion AC-YVAD-CMK worded through reducing the expression of caspase-l,IL-1β,IL-18 to effectively protect EAE rat from nerve damage.
