Relationship Between Transcription Factor 7-like 2 Gene And Type 2 Diabetes Mellitus Among Han Population In Xinjiang

Objective:To investigate the relationship between polymorphisms of transcription factor 7-like-2(TCF7L2) gene(rs11196218,rs10749127,rs290487,rs290481 and type 2 diabetes mellitus(T2DM) among Han population in Xinjiang. Methods:164 Han patients with T2DM who were treated in the First Affiliated Hospital of Xinjiang Medical University from November 2013 to February 2014,were included in T2DM group,at the same time,195 healthy people who received physical examination were included in control group. The height,body mass,abdominal circumference,blood pressure,levels of blood glucose.glycated hemoglobin(HbA1c),insulin,C-peptide,blood lipids and uric acid were measured and recorded. Peripheral blood DNA was extracted,genotype and allele frequency were detected for TCF7L2 gene rs11196218,rs10749127,rs290487,rs290481. The influence factors for T2DM were analysed by multivariable Logistic regression. Results:There was no significant difference in systolic blood pressure(SBP)between control group and T2DM group(P>0.05). The diastolic blood pressure(DBP),levels of three acyl glycerin(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C) fasting blood glucose and HbA1c of patients in T2DM group were significantly higher than those of patients in control group,but the high density lipoprotein cholesterol(HDL-C)level of patients in T2DM group was significantly lower than that of patients in control group(P<0.05). There was no significant difference in genotype and allele frequency distribution of rs11196218 and rs290481 between the control group and T2DM group(P>0. 05). There was significant difference in distribution of rs10749127 TT genotype between control group and T2DM group(P<0.05). There were significant differences in genotype and allele frequency distribution of rs290487 CC between control group and T2DM group(P<0.05). There were significant differences in levels of fasting glucose and uric acid among T2DM patients with different rs10749127 genotypes(CC,CT and TT) (P< 0.05). But there was no significant difference in various indexes among CC,CT and TT genotype of rs290487(P>0.05). According to multivariable Logistic regression analysis results,rs290487 CC genotype,abdominal circumference,BMI,levels of fasting blood glucose and HbA lc had correlation with T2DM(P<0.05). Conclusion:TCF7L2 gene rs10749127 and rs290487 are correlated with the onset of T2DM among Han population in Xinjiang,but the correlation between rs11196218,rs290481 and T2DM are not found.
