Objective:Constructing acute graft-verse-host disease (aGVHD) mouse model to explore whether the bone marrow dendritic cells treated by Galectin-1 (BM-DCgal) could prevent and treat aGVHD.Methods:(1) Obtain the bone marrow cells and Spleen lymphocytes from C57BL/6 mice as graft;(2) The bone marrow progenitor cells were prepared from BALB/c mice and then stimulated by GM-CSF, IL-4 and Galectin-1 to generate BM-DCgal, sort the BM-DCgal out by flow cytometry;(3) The mice of blank control group were injected PBS as graft after irradiating by gamma rays;(4) The mice of model group were injected bone marrow cells and Spleen lymphocytes from C57BL/6 mice as graft after irradiating by gamma rays;(5) The mice of intervention group were injected bone marrow cells and Spleen lymphocytes from C57BL/6 mice as graft and BM-DCgal as intervention programme after irradiating by gamma rays;(6) Observing and recording general state, life span and weight change of the recipient mice;(7) Peripheral blood from articulo mortis mice was collected to detect leucocyte count;(8) Post-mortem examination must be carried out immediately when mice died to collecte target organs of aGVHD which would be prepared into pathological sections and HE staining.Results:(1) The bone marrow cells and Spleen lymphocytes from C57BL/6 mice were prepared successfully;(2) The BM-DCgal were successfully prepared and sorted by flow cytometry;(3) The constructed aGVHD mouse model can be applied to the vivo study of prevention and treatment of aGVHD;(4) The morbidity of aGVHD among the mouse model which intervented with BM-DCgal was down-regulated;Conclusions:The BALB/c mice which injected bone marrow cells and spleen lymphocytes (spleen lymphocytes:5~*10~6, bone marrow cells 3~*10~7) from C57BL/6 mice as graft after irradiating by gamma rays may suffer from aGVHD, at the same time, diagnosis of aGVHD may be supported by the clinical and pathologic score of liver, spleen, lung, kidney, intestine and skin. The aGVHD morbidity of intervention group injected bone marrow cells and spleen lymphocytes(spleen lymphocytes:5~*10~6, bone marrow cells 3~*10~7) from C57BL/6 mice as graft and BM-DCgal (1~*10~6) as intervention programme after irradiating by gamma rays among the mice were down-regulated obviously. This study may provide experimental basis for the prevention and treatment of clinical aGVHD.
