Objective:To establish the animal model of small-diameter vascular prostheses transplantation.Test vascular prostheses patency rate in different time after transplantation. The study compared differences in preoperative and postoperative autologous blood vessels. Comprehensive analysis of the experimental methods and results. To make a scientific evaluation with vascular prostheses short-term patency rate.Of small diameter artificial vascular to make scientific evaluation.Methods:Replaced 16 beagle dogs right femoral artery with small-diameter vascular prostheses. Vascular prostheses with silk fibroin as material, the inner diameter 3 mm. All cases were randomly assigned to A group of 8 cases,8 cases of group B before surgery. Taken out natural vascular specimens for the self control group (mark with Ao, Bo) when vascular replacement surgery began. The day after surgery, postoperative 3 months, postoperative 6 months using color Doppler ultrasound to detect vascular patency rate. Collect A group specimens on the day of postoperative 3 months. Collect B group specimens on the day of postoperative 6 months. Using HE staining, Von Kossa staining and Masson staining for histological observation of blood vessels. And the vessel wall thickness measurement.Contrast the difference between groups. And the results were statistically analyzed.Results:1. Operation of vascular replacement success rate was 100%. Intraoperative observation found that vascular prostheses beat well. Vascular ultrasound found that all the vascular prostheses were without stricture the day after surgery. No animal death of intraoperative and postoperative death. No surgical incision infection.2. A group 3 months after surgery:ultrasonic testing patency rate was 75% (6/8). B group 3 months after surgery:ultrasonic testing patency rate was 75% (6/8), and six months after surgery:ultrasonic testing patency rate was 12.5%(1/8). Three months after surgery, two groups of vascular ultrasound of patency rate was same. B group vascular ultrasound results found that the patency rate with 6 months after surgery was less than the patency rate with 3 months after surgery. The difference was statistically significant (P< 0.05).3. HE staining:(1) the self control group:normal blood vessels form, complete structure, blood vessel wall stratification clear; (2) A group:blood vessel wall thickening, give priority to intimal thickening, wall stratification was clear; (3) B group:obvious thickening of the blood vessel walls, stratification was not clear. Calcification.4. Masson staining:(1) the self control group:normal vascular structures. Collagen fiber was stained in blue by aniline blue, mainly distributed in vascular smooth muscle layer and outer membrane of blood vessels. Collagen fibers and muscle fiber cross arranged in smooth muscle layer. (2) A group:endangium thickening, all layer of blood vessels was covered a large number of collagen fibers (blue).The compact structure of fiber; (3) B group:the blood vessel wall was not clear of stratification, and obvious thickening. Muscle fibers (red) from the previous increase, and arranged disorderly.5. Von Kossa staining:(1) the self control group:normal vascular wall structure. Vascular full-thickness visible in the black spots. No obvious calcium deposition. (2) A group:vascular wall thickening, each layer was not clear. Vascular full-thickness visible in the black spots. No obvious calcium deposition. (3) B group:obvious thickening of the blood vessel walls, deposition of calcification.6. Vessel wall thickness:self control group (A0):274.±10.46μm, A group: 381.25±30.94μm, self control group (B0):273.63±9.47μm, B group: 508.63±39.26μm. self control group (A0) compared with self control group (B0), there was no statistically significant difference(P> 0.05). Self control group (A0) compared with A group, the difference was statistically significant (P< 0.05). Self control group (B0) compared with group B, the difference was statistically significant (P< 0.05). A group compared with B group, the difference was statistically significant (P< 0.05).Conclusions:1. This experiment to establish animal models of success rate was 100%. Modeling method had some innovation.2. This experiment using small-diameter vascular prostheses patency rate 3 months after implantation was 75%, and the patency rate 6 months after implantation was 12.5%. The patency rate of postoperative 6 months was low. Vascular prostheses need to improve the performance.3. The small-diameter vascular prostheses stenosis and occlusion after implantation, hyperplasia of natural vessels of anastomotic stoma, may have close relationship with surgical operation, postoperative anticoagulation and materials of vascular prostheses.
