[Objective] To discuss the interrelation of CYP3A gene polymorphism and glucocorticoid osteoporosis in the han nationality in yunnan.[Method] Collecting the clinical data of 98patients treated with glucocorticoid(GC) and 91 volunteers in han nationality in yunnan.Based on the selected and eliminate standard, dividing into the glucocorticoid groud (GC group) and the control group (NC group).And using the dexa to test the density of lumbar spine.According to the Z value of bone density, the GC group can be divided into osteoporosis(OP) subgroup and non-osteoporosis subgroup.Extracting peripheral blood genomic DNA of 189 caces,by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method to measure the gene polymorphism of cytochrome P-450 (CYP) and the family of CYP3A4*18b(G20230A),CYP3A4*4(A13989G),CYP3A5*3 (A6989G), and analyze the the correlation of CYP3A4*18b, CYP3A4*4, CYP3A5*3 between the gene polymorphism and glucocorticoid induced osteoporosis (GIOP).[Result] 1、In GC group:①There was no statistically difference of BMD、the Z value between genotypes of CYP3A4*18b(G20230A)GG、GA、AA genes (P>0.05). Also,there was no statistically difference of BMD、the Z value between genotypes of CYP3A5*3 (A6989G)AA、AG、GG genes(P>0.05). ②In NOP sub group and OP sub group,the distribution difference of genotype and alelic gene which belong to CYP3A4*18b site and CYP3A5*3 site have no statistically significant(P>0.05).2、①In GC group, the average bone mineral density of L1-L4 is 1.036±0.157,the average bone mineral density of control group is 1.135±0.155,the difference is statistically significant(P<0.05); ②The ratio of osteoporosis in GC group(19.3%) is higher than the ratio in control group(0.03%),the difference is statistically significant (P=0.001); ③The time of taking GC in OP group is longer than NOP group,the difference is statistically significant(P<0.05)。3、In the experiment,CYP3A4*18b(G20230A)A allele frequency has statistical significance compared with Chang xinyu et al’s Liaoning Province Han healthy population CYP3A4*18b distribution differences(P<0.05),this may be related to regional differences.The allele frequency of CYP3A5*3(A6989G)G is 86.51%,it has statistical significance compared with Li dingyun et al’s distribution of CYP3A5*3 in Hunan Province Han healthy population(P<0.05),this may be related to the regional differences of Kunming.[Conclusion] 1.The CYP3A4* 18b(G20230A)A allele frequency、CYP3A5*3 (A6989G)G allele frequency of Han Chinese in Yunnan are higher than normal.2.The gene polymorphism of CYP3A4*18b(G20230A)、CYP3A5*3(A6989G) have nothing to do with the occurrence of osteoporosis after taking GC.3.The ratio of osteoporos of patients taking GC is higher than control group,and the average bone mineral density of L1-L4 is lower than the control group.The longer the time to take GC, the greater the injury.
