Objective:We evaluated the effects of Bufeiqingyu granules on the expression of Smad2、 Smad3、Smad7、connective tissue growth factor (CTGF) on bleomycin-induced scleroderma in rodent model,in order to provide the basis of in treating scleroderma by lungs.Methods:60 BALB/c mice,which were 8 weeks old femal,were randomly divided into five groups:blank control group,BLM model group, and low-, middle-, high-dose Bufeiqingyu granules groups.The model for scleroderma was replicated by continuous intradermal injection of bleomycin in back area once per day.Mice in blank group received continuous intradermal injection of physiological saline in back area once per day. The three dose groups of Bufeiqingyu granules began to take small, medium and large doses respectively, and the blank control group and model group were given clear water.After 28 days we put all mice to death,cut off the lung and back skin. Smad2、Smad3、Smad7、CTGF mRNA and protein were determined by fluorescence quantitative PCR and immunohistochemical(IHC) technique in the lung and skin tissue.Results:1.Compared with the blank group, model group of skin and lung tissue fibrosis, lung dredging particles and large dose groups also appeared fibrosis,but compared with model group,The degree is lighter than the model group.2.Compared with the model group,large dose of Bufeiqingyu granules can reduce the skin and lung tissue of Smad2 and CTGF protein expression, especially in high dose effect is obvious (**P<0.01), but have no obvious effect in Smad3 and Smad7.3. Compared with the model group,large dose of Bufeiqingyu granules can reduce the skin and lung tissue of Smad2 and CTGF mRNA expression, especially in high dose effect is obvious (**P<0.01), but have no obvious effect in Smad3 and Smad7.Conclusion:Bufeiqingyu granules could significantly inhibit the high expression of Smad2、 CTGF mRNA and protein in the lung and skin tissue,but have no obvious effsct in Smad3 and stimulating effect I-Smads such as Smad7.So we supposed that Bufeiqingyu granules regulated the metabolism of CTGF by inhibiting the expression of Smad2,and then improve the pathological injury of scleroderma, prevent the progress of skin and organ fibrosis.
