Study Of Longchai Formula In Sheldrake Models On HBV Viral Infection Inhibition

In this study,we summary and analyze the theoretical literature and the status of Chinese and Western medicine research on CHB and carry out the experimental study with the establishment of DHBV duck infection model, observe the effect of changes in serum virus content of animal models by Longchai formula’s continuing role for explore its anti-viral DHBV inhibition,provide scientific basis for clinical efficacy.Part I:Theoretical study We discusse from the modern medcine’s etiol-ogy..epidemiology,diagnosis and treatment,drug evaluation on CHB and the overview of the research related to it;besides,the theoretical analysis of the role of the efficacy on CHB of Longchai formula are discussed.Theoretical studies suggest that modern medicine for HBV infection transmission routes and mechanisms are clear,nucleoside analogues (NAs) and interferon (IFN) anti-HBV effective and objective,but persistent resistance,side effects and withdrawal after a rebound even worse become the focus the major problem.Chinese medicine believes CHB pathogenesis and pathological factors wet,hot,melancholy,stasis,drugs,and other related pathological factors,and mainly dialectical type of treatment,effective experience formula,integrative medicine treatment and other methods,has made gratifying achievements.Longchai formula on CHB objectively and effectively,has a certain anti-HBV effect,its efficacy is based on the role of traditional Chinese medicine theory and based on efficacy and its prescrip-tion drug Pharmacology mechanisms related to,but still need to test their anti-HBV effect research and evaluation.Part II:Experimental Study Establishing the duck HBV infection model,through Longchai formula’s high, medium and low dose interven-tion,contradistinguish with positive drug lamivudine(3TC)administration and the virus contorl,observe Longchai formula of HBV infection inhibition of tne infected duck’s serum DHBV DNA. The results showed that,①each drug intervention group at D0,D10 days DHBV DNA expression was significantly decreased with virus con-trol(P<0.01);②lamivudine pre inhibition of DHBV DNA is better than that of Long-chai formula groups(P<0.05and0.01),but after stopping,inhibition decreased obvi-ously and DHBV rebound,while each group of Longchai formula showed continuous inhibition;③between Longchai formula high,medium and low dose group compared to the low dose group best inhibitory effect,inhibition continued;④Long Chai Fang high,medium and low dose groups did not exhibit inhibitory effect on DHBV clear dose-response relationship. Analysis,Longchai formula on DHBV inhibited,although the short-term effects of its anti-HBV compared with lamivudine is low, but it shows long-lasting anti-HBV effects;as for the high dose did not show better than low the role of anti-HBV activity dose group,consider the possible toxic effects of large doses of prescription drugs nightshade,hedyotis diffusa of cell-associated,so that the cellular immune function reduced,HBV active proliferation increased.This study suggests that the clinical treatment of CHB, in the use of NAs and IFN same time,with the use of conventional doses of Longchai formula will likely improve the anti-HBV ef-fect,lasting immune response and improve efficacy;while avoiding the use of high-dose prescription.This study provide Longchai formula has the role of anti-HBV experimental basis, is also provide scientific evidence of traditional Chinese medicine efficacy on CHB.
