Purpose:To investigate the effect and mechanism of traditional Chinese medicine of QingganZishen by upregulation of SIRT1 expression in visceral adipose tissue to improve hypertension metabolic disorders.Methods:To get spontaneously hypertensive rat model of metabolic disorders induced by high-fat diet.17 4-week-old male spontaneously hypertensive rats (SHR) were chosed. All rats were divided into high-fat diet group 12 and normal diet group 5. They were respectively given high-fat feed and basal feed. The high-fat diet group was randomly divided into the Zhenjian granules group 6 (22.7g/kg/d, filling the stomach) and the model control group 6 (same volume distilled water, filling the stomach). Rats were weighed twice weekly, but also blood pressure. Then we changed the amount of drug based on their body weight. Rats were sacrificed after administration for 9 weeks, the visceral fat (epididymis) was acquired and weighed. Apart of the visceral fat was used to separate the fat cells, a part was used to extract RNA and protein, a part was fixed with 4% formaldehyde for HE staining.1)We evaluated the metabolism of SHR through regular monitoring blood pressure and body weight;2)By RT-PCR and Western Blot, we assay Sirtl mRNA and protein levels, and cleavage enzyme caspase-1 protein levels in visceral adipose tissue, and evaluate Sirtl expression level and activity;Results:1.The influence of Chinese medicine of QingganZishen on body weight in SHR rats:No significant difference in body weight between the three groups of SHR rats at baseline(P>0.05). After 9 weeks of intervention, the body weight of model group was significantly higher than the control group(P<0.01), and it of the Zhenjian particles group was significantly lower than the model group(P<0.01). There was no significant difference between the control group and the group of Zhenjian particles.2.The influence of Chinese medicine of QingganZishen on blood pressure of SHR rats: Before treatment, the SBP of model group was higher than that of the control group (P<0.05), and there was no significant difference between the other groups (P>0.05). There was no significant difference between the three groups in DBP(P>0.05). After 9 weeks of intervention, there was no significant difference in blood pressure between the model group and the control group. Although there was a downward trend in the needle and arrow group, there was no statistical difference.3.The influence of Chinese medicine of QingganZishen on the changes in volume of visceral fat and content of lipid droplets in SHR rats:3.1 The comparison of the fat weight around epididymal:a. After 9 weeks of intervention, the fat content of the model group was higher than that in the control group (P< 0.05), but there was no significant difference between the model group and the Zhenjian particles group(P> 0.05);b. The fat ratio of model group was significantly higher than that of control group (P< 0.01), but there was no significant difference between the model group and the Zhenjian particles group(P>0.05).3.2 Assessment of fat mass and lipid droplets in the epididymis under the microscope: Compared with the control group, the size of adipocytrs of the model group and the lipid droplet content increased; the size of adipocytrs and the lipid droplet content decreased compared with the model group.4. The influence of Chinese medicine of QingganZishen on the changes of SIRT1 expression and activity in visceral adipose tissue:4.1 The influence on the expression of SIRT1 mRNA in adipose tissue around epididymal:After 9 weeks of intervention, the expression of SIRT1 mRNA in the model group was significantly lower than in the control group (P<0.01), and it in the Zhenjian particle group was significantly higher than in the model group (P<0.05).4.2 The influence on the expression of SIRT1 protein in adipose tissue around epididymal:After 9 weeks of intervention, the expression of SERT1 protein in the model group was significantly lower than in the control group (P<0.01), it in the Zhenjian particles group was significantly higher than in the model group (P<0.05).4.3 The influence on the expression of Caspasel protein in adipose tissue around epididymal:After 9 weeks of intervention, the expression of Caspase 1 protein among the three groups was no significant difference (P> 0.05).Conclusion:1.Traditional Chinese medicine of Qingganzishen (Zhenjian granules â…¡) can reduce hypertensive rats complicated with metabolic disorders with body weight, reduce visceral fat accumulation.2.Traditional Chinese medicine of Qingganzishen (Zhenjian granules â…¡) may up-regulate the expression of SIRT1 in visceral adipose tissue to improve the metabolic disorder of hypertension.
