Background & Aim:In human malignant tumors, enhanced glycolysis is a common feature. The increased expression of glucose transporters of glucose utilization and metabolism of tumor tissue is closely related to the increased transport capacity strong isoforms of glucose transporter protein 1 (Glut-1) is the key to promote the synthesis of increasing its role. Protein through the activation of the Glut-1 gene, which mediates the uptake of glucose increased, the energy required to meet the tumor, but also for the invasion, metastasis, recurrence and provide corresponding conditions. Recent studies show that the expression of Glut-1 protein in gastric cancer tissues than in normal tissues is different. Therefore, this study aims to explore the mRNA expression level of Glut-1 in human gastric cancer tissue and normal gastric mucosa and relationship with clinicopathological features of gastric cancer patients.Methods:In this study, we selected patients who underwent surgical resection in 61 cases of gastric carcinoma and corresponding normal tissue samples (from the cut edge of>5cm) as the experimental group and the control group respectively in Clinical Medical College of Yangzhou University(Jiangsu Subei People’s Hospital) from June 2014 to June 2015. None of the patients received the chemical therapy or radiotherapy before resection, which are proved by pathology. Under the condition of low temperature liquid nitrogen grinding. Trizol method for extracting total RNA in all samples, the application of reverse transcription the total RNA reverse transcription kit cDNA, application of RT-PCR (Real-time Polymerase Chain Reaction) expression method for quantitative detection of Glut-1 mRNA, with three glyceraldehyde-3-phosphate dehydrogenase gene (hGAPDH) as the internal reference. At the same time using the expression of immunohistochemical staining of Glut-1 protein was detected, and the clinical and pathological features of Glut-1 patients with mRNA expression level with gastric cancer were analyzed.Results:Glut-1 mRNA in gastric cancer tissues at well, mediate and poor differentiated adenocarcinoma relative expression amount is 2.68±3.66、6.97±25.79、41.31±28.07 respectively. Three groups of difference has statistical significance (P<0.05); Pairwise comparison and differentiation degree is poor, Glut-1 relative expression quantity is high. The expression level of mRNA Glut-1 in the lymph node metastasis group (21.39±28.28) was higher than that in the non lymph node metastasis group (3.66±5.34), and there was significant difference between the two groups (P<0.05).Glut-1 mRNA in gastric cancer tissues in phase Ⅰ, phase Ⅱ, phase Ⅲ and phase Ⅳ adenocarcinoma of the relative expression quantity were 2.03±3.35、4.09±6.15、11.89±16.57、 40.29±40.44 and four group difference has statistical significance (P<0.05); expression level Ⅲ-Ⅳ group was significantly higher than that of group Ⅰ and Ⅱ reached level (P<0.05). The expression level of mRNA Glut-1 in signet ring cell gastric cancer tissues (2.12±4.97) was significantly lower than that in non-signet ring cell cancer group (13.47±22.61), and there was significant difference between the two groups (P<0.05). The expression level of mRNA Glut-1 in gastric cancer was positively correlated with tumor size, and the difference was statistically significant (r=0.529, P<0.05); and PLR was negatively correlated (r=-0.242), but there was no significant difference (P=0.06). Gastric cancer Glut-1 protein negative, positive, strong positive expression rates were 18.3%,44.6%,37.1%. The expression level of mRNA Glut-1 in gastric cancer was not related to gender, age, morphological classification, invasion degree, and vascular invasion.Conclusion:The expression level of Glut-1 gene was consistent with the level of protein expression. The high expression of Glut-1 gene may play an important role in the occurrence and development of gastric cancer. May be related to the invasive index of gastric cancer; Glut-1 may as indicators of prognosis in patients with gastric cancer analysis. Signet ring cell cancer genes Glut-1 expression may decrease its special cell structure, pending further study.
