ObjectiveBuPishen Huoxie traditional Chinese medicine developed by professor Wang Xiaoguang can tonify kidney and spleen, promote blood flow, ascend clear and dispel turbidity in Chronic renal failure. Clinical practice proved that the CRF patients using this medicine can achieve good curative effect.This study observed the effect of BUPISHENHUOXUE decoction on the expression of PCNA, Bcl-2 and i-PTH levels in the renal hyperparathy-roidism rats, and explored the regulation of parathyroid glands cell apoptosis and proliferation, to provide experimental basis for the further application.Methods5/6-nephrectomized (Nx) and sham operated with high phosphorus diets SD male rats were divided into five Groups (ten rats in each group):Sham-operated group(Sham),5/6nephrectomizedgroup(Nx),5/6-nephrectomized+calcitriolgroup (Cal),5/6-nephrectomized+traditional chinese medicine group (Tcm),5/6-nephrectomized+calcitriol+traditional Chinese medicine group (Tcm+Cal). Each group of rats were fed with adaptive normal diet after a week, then high phosphorus for 2 weeks before getting 5/6 nephrectomy, finally continued feeding of high phosphorus until death in rats. Blood samples were collected from the orbital vein at one week before the operation and respectively at the 1st,2nd,3rd,4th week after the operation. Serum phosphorus, i-PTH, BUN/SCr and other indicators were detected. At the end of the experiment (4 weeks after the operation), thyroid and parathyroid complexes were removed for pathological examination, to observe the expression of PCNA and Bcl-2 in the parathyroid gland of the rats.We compared the number of parathyroid gland cell per unit area and the expression of the apoptotic marker TUNEL by immunohistochemistry.Analyzed the exact effect of Chinese and western medicine combination through the inter-group control between groups of Tcm group, Cal group and Tcm+Cal group to provide theoretical basis for clinical application.ResultsAfter 5/6 nephrectomy & high phosphorus feeding, serum levels of SCr, BUN, P, iPTH of rats were significantly higher than those Sham group (P<0.05); the volum of parathyroid glands of high phosphorus treatment rats got bigger obviously, PCNA and Bcl-2 expression level increased sharply. Tradictional Chinese medicine intervention treatment group can significantly reduce secondary hyperparathyroidism rat serum creatinine, phosphorus, parathyroid hormone and other biochemical markers (P<0.05), but no significant effect on the improvement of the treatment groups BUN levels, and in calcitriol to improve biochemical indicators and no less pure Tradictional Chinese medicine treatment has obvious advantages. In addition, compared to traditional Chinese medicine intervention, calcitriol therapy alone can reduce PTH levels can not effectively improve hyperphosphatemia and renal function. Meanwhile, Tcm+Cal can effectively reduce the expression of parathyroid glands PCNA and apoptosis related protein Bcl-2 levels (P<0.05), more significant than the effect of traditional Chinese medicine group and the calcitriol group and medicine group and three calcium alcohol group in this respect there is no significant difference in efficacy between.ConclusionBUPISHENHUOXUE decoction can reduce SHPT rats’serum level of SCr, BUN, P, iPTH and inhibit excessive proliferation of rats’parathyroid gland as well as induce the cell apoptosis of proliferative parathyroid gland, improve the general situation of rats, indicating that BUPISHENHUOXUE decoction is an effective Chinese materia medica preparation for preventing and delaying the progress of the chronic renal failure sourced SHPT.
