ObjectiveThrough the observation of Qingre Toubiao Recipe (QTR) preventive treatment to mice infected with Influenza Virus FM1, using the important pattern recognition Toll-like receptor (TLR) 7 within innate immune response as the research target, we focus on discussing the QTR mechanism on fighting against the Influenza Virus. So as to provide important evidence for Chinese medicine’s anti-Influenza Virus theory research and clinic practise.Methods1) Using the nasal approach to build the model of mice with Influenza Virus caused lung injury, we observed the medical treatment’s death protection index and lung index. We also utilizing HE staining to observe the pathological changes within the mice lungs, so as to evaluate the medicine’s in-body anti-Influenza Virus effects2) Using the method of ELISA test to detect the content of IL-1β、TNF-α 、IL-6 within the serum of Influenza Virus caused lung injury mice, so as to evaluate the medicine’s ibitory effect to inflammatory factor after influenza virus affection3) Using the Fluorescence quantitative PCR and western-blot methods to detect the TLR7、MyD88、NF-κB、AP-1mRNA and protein expression level within the Influenza Virus caused lung injury mice, and evaluate the medicine’s influence to the TLR7 mediated signal pathway within the Influenza Virus infected lungResults1) Compared to the model group, the high and middle dose QTR group could effectively reduce the mice death rate, increase the death protective rate, and improve living conditions. Their difference is statistically valid (P <0.01或<0.05). The low dose QTR group did not show clear protection against death. It may caused by low medicine dosage. All QTR dose groups could lower the influenza virus infected mice lung index compared to the model group. Their difference is statistically valid (P<0.01 或 P<0.05). In microscopically, the recipe can bring various improvement to the influenza virus caused virus lung injuries and reduce Lymph and other inflammatory cells, easing the infiltration, and reduce the congetion of alveolar2) Compared to the model group, all QTR dose groups can effectively lower the levels of IL-1β、TNF-α、IL-6 within the serums of the influenza virus infected mices. The difference is statistically valid (P<0.01 或 P<0.05) It shows that the recipe can effectively reduce the levels of pro inflammatory factors such as IL-1 β、TNF-α、IL-6, avoiding the Immunological injury caused by excessive expression of inflammatory cells3) the high and middle dose QTR group could lower the expression of TLR7、 MyD88、NF-κB、AP-1 mRNA and proteins. Compared to the model group, it is statistically valid (P<0.01 或 P<0.05). The low QTR dosage group did not show clear difference (P>0.05) to the model group. It means the recipe’s high and middle dosage can suppress the inflammatory cell factor expression by lowering the TLR7 mediated MyD88 dependent signal transduction pathway, so as to show the antiviral effectConclusionThe QTR can effectively suppress the proliferation of influenza virus FM1 within the mice body, greatly reduce the release of inflammatory factor, and reduce the inflammatory pathological lesion of lung. Its anti-influenza virus mechanism could be suppressing the inflammatory cell factor expression by lowering the TLR7 mediated MyD88 dependent signal transduction pathway...
